Abstract
Bu buluş gıda, su, tarım, tıp, biyoloji, kimya ve bunun gibi alanlarda kullanılan ve triptofan aminoasidini ayrıştırarak indol meydana getirme yeteneğine sahip bakterilerin kontaminasyonundan kaynaklanan toksik niteliklerin belirlenmesini sağlayan bir hijyen tespit cihazı (1) ile ilgildiir. E. Coli ve bunun gibi bir bakterinin ürettiği triptofanaz enzimi ile triptofan etkileşime girmekte ve deaminasyon ile triptofandan purivik asit, amonyak ve indol meydana gelmektedir. Buluş konusu hijyen tespit cihazında (1) öncelikle bakteriye 37 derecelik bir etüv ortamı sağlanmakta ve 4-8 saat boyunca bakteri 37 derecede inkübasyona bırakılmaktadır. 4-8 saat sonunda gelen uyarı ile bakteri tarafından yeterli miktarda oluşturulan indol üzerine 0.5 ml Kovaks reaktifi damlatılmaktadır. Kovaks reaktifi izoamil alkol, paradimetilaminobenzaldehit ve hidroclorik asitten oluşan biyokimyasal bir reaktiftir. Bakteri bakteri tarafından üretilen indol para-dimetilaminobenzaldehit ile etkileşmekte ve bulundukları ependorf yüzeyinde vişne çürüğü renginde bir tabaka oluşturmaktadır. Optik okuyucu (6) bu vişne çürüğü renginin frekansı algılamakta ve varsa bakteri varlığı LCD ekranda (8) kullanıcıya aktarılmaktadır.
