------04/12/2563------(OCR) 1. กรรมวิธีการตรวจหาเชื้อก่อโรคในอาหารด้วยแอนติบอดีอะเรย์ที่มีลักษณะเฉพาะคือ สามารถตรวจหาเชื้อก่อ โรคในอาหารตั้งแต่ 1 ชนิดขึ้นไปได้พร้อมกันในการตรวจครั้งเดียว ซึ่งประกอบด้วยขั้นตอน ดังนี้ 1) การตรึงแอนติบอดีปฐมภูมิบนแผ่นสไลด์ (spotting); The method for detecting foodborne pathogens using a unique antibody array allows for the simultaneous detection of one or more foodborne pathogens in a single test. The process consists of the following steps: 1) Primary antibody immobilization (spotting): Primary antibodies specific to the foodborne pathogen are dissolved in a suitable buffer for immobilization, such as phosphate-buffered saline (PBS) pH 7.2, and then immobilized (spotted) onto a slide coated with poly-L-lysine or another coating substance at a volume of 0.5 microliters per drop (spot). The slides are then stored in a humidity-controlled cabinet at 30-35 percent humidity and 23-25 degrees Celsius for approximately 12-16 hours. 2) Preventing nonspecific reactions (blocking): This is achieved by adding a suitable buffer solution. To prevent nonspecific reactions on the slide surface, use a 3% (weight/volume) buffer solution of skim milk in phosphate buffer saline (PBS, blocking solution) on the slide surface. Let it sit at room temperature for at least 15 minutes. 3) Washing the slide: Wash the slide with a suitable buffer solution for washing, such as phosphate buffer saline (PBS) containing 0.05% Tween20, three times for 3-5 minutes each time, gently shaking and rinsing. To store the slide for testing, dry it and store it at 4°C. It can be stored for at least 2 months. 4) Place 100 µl of the sample solution containing bacteria dissolved in 1% (weight/volume) skim milk on the slide surface and cover with a glass slide.
