Description
The invention relates to the microbiological industry and is intended to control the activity of Actinomyces grisens biomass (liquid culture) at various stages of the production process. There is a method of controlling the activity of biomass Actinomyces grisens 15, which involves measuring the optical DENSITY of the colored solution obtained as a result of the preliminary chemical treatment of the native solution on the FEC at a green optical filter 7. - 540 MMK and determining the activity of the biomass by a calibration curve depending on the average of the optical density of the obtained colored solutions of Å. A disadvantage of this method is the need to chemically treat a native biomass solution. s before measuring its optical density, which complicates the control and reduces its degree of accuracy. The purpose of the invention is to accelerate and improve the accuracy of the analysis. This goal is achieved by the fact that the optical densities of the native solution are measured at characteristic absorption bands 1055 and V. 1660, and the activity of the biomass is determined by the ratio of the measured optical densities. The drawing shows the calibration curve of the dependence of the ratio of optical density and biomass activity. Example. Take 20 ml of the culture liquid under study, acidify with pH 2.5 to pH 2.5, hold for 20 minutes and get the native solution by filtration through filter paper. 1.5 ML of the native solution is pipetted onto a 40 mm diameter CaF substrate, bounded by a ring of the same diameter. To remove moisture, place the substrate in an oven at 370 s. After the formation of a thin film of dry matter, the optical density on IKS-22 for I, 1055 cm and J ° is calculated, the ratio of measured optical densities on characteristic absorption bands is measured, and then the biomass activity is determined by the calibration curve of the dependence of optical ratio
