Description

Изобретение относитс  к области искусственного осеменени  животных. Известен снособ подобного назначени , включающий смешивание спермы с флуоресцентным индикатором и ее оценку под микроскопом . Однако он требует значительной затраты времени, так как основан на определении качества спермы по соотношению живых и м,ертвых сперматозоидов. С целью ускорени  и повышени  точности определени  в предлагаемом способе после смешивани  спермы с флуоресцентным индикатором измер ют врем  изменени  флуоресценции спермиев, по которому суд т о качестве спермы. Способ осуществл етс  следующим образом . На предметное стекло нанос т каплю спер мы и каплю флуоресцентного индикатора, например , 0,025-0,03%-ного раствора акридинового оранжевого, приготовленного на 2,8%ном растворе лимоннокислого натри  (трехзамещенного п тиводного) и 1%-ной двууглекислой соды. Обе капли смешивают, покрывают покровным стеклом и помещают на предметный столик микроскопа, использу  дл  освещени  светофильтры: 2СЗС-21, 2ФС-1 и 2БС-3, а на окул рах микроскопа - Л ;С-18 или ЖС-19. При микроокопировании спермы живые спермии флуоресцируют красным цветом, а мертвые- рко-зеленым. В процессе наблюдени  красна  флуоресценци  замен етс  зеленой . Причем в одних эокул тах эта смена цветов происходит быстро, в других - медленио . Врем  изменени  флуоресценции спермиев определ ют с помощью секундомера, и по скорости изменени  окраски оценивают оплодотвор ющую снособиость да.ННОго эокул та - чем медленее смен етс  окраска, тем выше опло-дотвор юща  способность спермы. Пример. На экспериментальной базе «Заречье Белорусского научно-исследовательского института было исследовано 94 эокул та хр ков. Наблюдени , за оплодотвор емостью свиноматок ноказали, что если врем  изменени  флуоресценции спермиев менее 3 мин, то оплодотвор емость составл ет 54,5%; 3-4 мин -70,3%, более 4 мин - 86,8%.
