Description
The invention relates to the field of medicine and concerns methods for the production of purified enzymes, namely cytochrome C, which can be used as a therapeutic drug.
A known method for producing cytochrome C by extracting animal raw materials in an acidic medium, followed by adsorption and elution of the target product.
This method does not provide a satisfactory yield of cytochrome C and results in a product of insufficient quality.
The aim of the invention is to increase the yield and improve the quality of cytochrome C.
For this, the first stage of raw material extraction is carried out at a neutral pH of 7.0-7.5. Next, the process is conducted by well-known techniques.
Example. 10 / cg of fresh beef hearts are crushed and 10 liters of water containing 0.5 g of ethylenediaminetetraacetic acid are poured. Adding 0.1 n. The caustic soda solution is adjusted to pH 7.5, stirred and squeezed out on a press after 15 minutes. To the residue was added 10 l of water with ethylenediaminetetraacetic acid, and the pH of the mixture was adjusted to 5.0 with a concentrated solution of citric acid. After two hours of non-stirring at room temperature, the mixture is pressed again to the press and poured onto the resulting extract with 0.1N. caustic soda solution to pH 7.5. The precipitated precipitate is separated by filtration, and amberlite is added to the filtrate with stirring in an amount of 10 g of dry resin to 1 l of extract. The mixture is stirred for 10-15 h, settled for 1-2 hours, the solution is drained, and the precipitate is washed by decantation with distilled water. The washed ion exchanger with adsorbed cytochrome C is treated for 10 minutes with a solution of sodium chloride, taken in an amount of 2.5% of the volume of the initial extract. The eluate is dialyzed and then fractionated with ammonium sulfate at pH 7.5 within 80-100% of saturation. The fraction collected after centrifugation at 6000 rpm is dissolved in 50 ml of water and dialyzed. After dialysis, the solution is lyophilized. The yield is 2.5 g of dry cytochrome C.
Subject invention
The method of obtaining cytochrome C by extracting the crude animal with water at an acidic pH value, followed by adsorption and elution of the target product, characterized in that, in order to increase the yield of cytochrome C and improve its quality, the raw material is pre-extracted at neutral pH and then the process is famous tricks.
