This invention relates to biotechnology and relates to the production of bacterial starters for biological preservation of feed.
The aim of the invention is to increase the biomass yield and simplify the technology.
The method is carried out as follows.
A nutrient medium suitable for growing both cultures is prepared, seeded with both cultures, co-cultivated under optimal conditions at 31-32 ° C and upon completion of the cultivation process, the resulting biomass is concentrated, mixed with protective medium and dried.
The duration of the cultivation process does not change and on average is
22 ч. 22 h. As a result of co-cultivation, the ratio of cells of Streptococcus lactls dlastatlcus was 20 IMV of the Academy of Sciences of the Kazakh SSR and Proplonlbacterlum shermanli pcs. 12 The IMV of the Academy of Sciences of the Kazakh SSR is from 1.0-1.0 to 1.0: 1.4, which meets the regulatory requirements. In other words, the adjustment of the bacterial composition of the biological preparation is not required. The accumulated mixed bacterial biomass is concentrated, mixed with a protective medium and dried in a known manner. In accordance with the current regulations, drying can be carried out by both sublimation and low-temperature spraying. In tab. 1 shows the yield of bacterial biomass of lactic acid bacteria in the implementation of the proposed method.

2.When implementing the proposed method, the cell ratios of the two cultures lie within the requirements of the technical conditions for the final product only in the case of co-cultivation of crops at 31-32 ° C, and at temperatures of 29 and 36 ° C, at which the respective cultures are separately cultivated, the required ratio no cells are provided. Relevant data is given in Table. 2
The use of the proposed method allows to increase the yield of bacterial biomass at the cultivation stage, increase the utilization rate of the nutrient substrate, greatly simplify the technology for obtaining bacterial starter culture by combining the cultivation processes of both bacterial cultures and eliminating the need to standardize the finished product according to the cell viability ratio of each of the cultures.
Example 1. The apparatus with a volume of 250 liters was filled with 200 liters of nutrient medium containing, wt.%: Corn starch 4.0; corn extract 2.5 (in terms of absolutely dry weight); ammonium sulfate 0.3; cobalt chloride 0.001; manganese sulfuric acid 0.016, water. After sterilization, the nutrient medium contained reducing substances of 0.2%, amino nitrogen 49 mg%, had a pH of 6.9. The medium was cultured on a material comprising 0.8 l of bacterial suspension of Streptococcus lactls diastatlcus (microbial cell content of 1.10-109 cells / ml) and 1.6 l of bacterial suspension of Propionlbacterlum shermanli (microbial cell content of 1, 26 109 cells / ml) .

Streptococcus lactlsCultivation lasted 22 hours at 31 ° C. The total biomass yield at the cultivation stage was 1.28 10 cells / ml. Streptococcus lactls cell output
1:1.diastatlcus was 0.6 -10 cells / ml, and Propionlbacterum cells. shermanli 0.68 109 cells / ml. The ratio of Streptococcus lactls diastatlcus and Propionlbacterlum shermanli cells was 1: 1.
Propionlbacterlum shermanliThe culture fluid was concentrated by supercentrifugation. The obtained wet biomass in the amount of 4.1 kg was stirred in 4.1 l of protective medium and dried using low-temperature spray drying. A total of 1.06 kg of bacterial preparation containing viable Streptococcum lactls shermanli cells 16 109 cells / ml and Propionlbacterlum shermanli were obtained
18 -10 cells / ml. Thus, 3.2 kg of the finished product that meets all the requirements of GOST was obtained.
