Abstract
Изобретение относитс к методам биохимического анализа, в частности к способам определени концентрации меланиновых пигментов. Цель изобретени - повышение точности способа. Количество меланина определ ют по степени ингиби- ровани реакции восстановлени нитроси- него тетразоли в формаэан в слабощелочной среде рН 8,2 в присутствии феназинметосульфата и восстановленного никотинамидадениндинуклеотида - НАДНа, или по ускорению реакции автоокислени адреналина в щелочной среде рН 10,2. Количество меланина определ ют спектрофотометрически по изменению начальной скорости образовани продукта реакций по сравнению с контролем. (В первом случае - по торможению начальной скорости образовани формазана при 560 нм во втором - по ускорению начальной скорости образовани адренохрома при 500 нм). 3 табл.This invention relates to methods for biochemical analysis, in particular to methods for determining the concentration of melanin pigments. Непосредственно перед спектрофотометрированием в реакционную смесь добавл ют 384 мкМ (1 мл) восстановленного никотинамидадениндинуклеотида - НАДН2. Общий объем реакционной смеси равен 3 мл. Контролем служит проба, не содержаща меланина.
