Description
The invention relates to the microbiological industry, in particular to a method for preparing a nutrient substrate based on a hydrolyzate of plant raw materials for growing yeast.
The purpose of the invention is to increase the yield of yeast.
The method is carried out as follows.
One of the stages of preparation of the nutrient substrate from the hydrolyzate of plant raw materials is purging with steam, which is subjected to the hydrolyzate before or after treatment in a known manner, including neutralization, crystallization and release of gypsum. After cooling, the vapor forms furfural-containing condensate, in which the concentration of furfural is determined, after which the condensate is treated with ozone, the amount of ozone being 0.2-0.3 of the amount of furfural in the condensate.
The clarified hydrolyzate is mixed with the ozone-treated condensate in the ratio l: (0.05-l) and used as a substrate for growing yeast. During the ozonation of furfurolsoda of condensing condensate, furfural is deeply oxidized with the formation of volatile organic acids (LOK), which serve as an additional carbon food for yeast.
Example 1. The hydrolyzate obtained by hydrolysis of a mixture of needles and hardwood at 180 ° C was blown with steam at 100 ° C for 2 hours. Furfural containing steam was condensed. The amount of condensate was 50% of the amount of blown hydrolyzate. Condensate with a furfural concentration of 0.11% and LOK 0.085% were treated with ozone at doses of 0.15; 0.22; 0.33; 0.4 g / l.
Осветленный нейтрализат доней- трализовывали аммиачной водой до рН 4,2, аэрировали при 80°С в течение 2ч, охлаждали до 40°С и отдел ли взвешенные частицы путем отстаивани .Purified from furfural, the hydrolyzates were neutralized with lime milk to pH 3.5 and the gypsum was crystallized at 90 ° C for 1 hour. The gypsum was separated by settling. The clarified neutralizer was neutralized with ammonia water to pH 4.2, aerated at 80 ° C for 2 hours, cooled to 40 ° C, and suspended particles were separated by settling.
I.The prepared neutralite was mixed with ozonated condensate, mineral salts and Candida Scotti yeast were added at 38-39 ° C. The results are presented in table. I.
0,24%.Example 2. The wood hydrolyzate was neutralized and gypsum was separated as in Example 1 and a cooling unit was applied to the vacuum, where the neutralized agent was cooled to 40 ° C. Self-evaporation vapors from the vacuum-evaporator unit were condensed and mixed with the condensate vapor obtained from the evaporators upon cooling the hydrolyzate from 180 to 100 C. The amount of condensate was 15% of the amount of hydrolyzate. The concentration of furfural in the condensate is 0.3%, the concentration of LOK is 0.24%.
дел ли взвешенные вещества путем отстаивани и смешивали с конденсатом , который предварительно озониро , 10After removal of furfural, the neutralization product was aerated for 1 h, the suspended substances were separated by settling and mixed with the condensate, which was previously ozonated, 10

1,3%.Example 1. The wood hydrolyzate was neutralized and gypsum was isolated as in Example 1 and was fed to a vacuum distillation column with a bottom heat exchanger. The vapors after the vacuum distillation column were condensed. The amount of condensate was 5% of the amount of neutralized. The concentration of furfural in the condensate is 2.0%, the concentration of LOK is 1.3%.
