Abstract

Изобретение относится к биотехнологии, в частности к генетической инженерии, и позволяет получить рекомбинантный вектор - аденовирус CELO/pUC 19. С целью упрощения способа встраивания ДНК в геном аденовируса проводят гидролиз рестриктазой HbaI ДНК генома аденовируса CELO и плазмидной ДНК и неполный гидролиз рестриктазой Bgli ДНК генома аденовируса CELO затем смешивают Bgli фрагменты ДНК, содержащие 39% левой части генома аденовируса и два Xbal-фрагмента аденовируса CELO, содержащие 33-100% генома аденовируса CELO, которые предварительно последовательно лигированы в XbaI сайте на 94% генома с линейной плазмидной ДНК pUC 19 смесью, указанных фрагментов проводят трансфекцию клеток аллантоисной оболочки куриных эмбрионов. В результате рекомбинации in vivo в куриных эмбрионах из двух перекрывающихся фрагментов образуется полноценный геном Ad CELO содержащий в несущественной области гетерологическую ДНК - плазмиду pUC 9. 2 с.п. ф-лы.
