This invention relates to biotechnology and may be used in virology.
The purpose of the invention is to simplify the process of cultivating viruses and increasing their titers.
Example 1. Tubular organs, such as the intestine, are cut into segments of equal length, usually corresponding to the length of the holders. Intestinal segmenia are washed three times in Hanks' solution with antibiotics. Then, one end of the intestinal segment is tied up with silk or catgut. The transected end of the intestine is brought to the holder, which has been strengthened in the lid of the culture vessel, and is pulled on it, while simultaneously turning the mucous membrane out of the stocking. The cheekbone end of the intestine is cut off with scissors.
При вращении роллерного культурного сосуда сегменты кишечника на держател х последовательно погружаютс в питательную среду и подвергаютс аэрации воздухом ,The intestinal segments so fastened on the holders are washed three times by immersion in cylindrical vessels with Hanks solution and transferred to a roller culture vessel, where, after sealing the lids, the nutrient medium is poured through the nipples to maintain the vitality of the bowel segments. During the rotation of the roller culture vessel, the intestinal segments on the holders are successively immersed in the nutrient medium and subjected to aeration with air,
П р и м е р 2. PRI mme R 2. Cultivation of diarrhea virus strain Oregon with 24V for growing tissue of the intestine according to the proposed method. Selection, preparation, and planting of the intestinal segments on holders is carried out as in Example 1. After sealing the vessel, nutrient medium without serum is introduced through the side fitting, but diarrhea virus is contained at a rate of 0.1 ml per ml of medium. The culture vessel is transferred to a roller. The reproduction of diarrhea virus in intestinal tissue is monitored by the results of the cytopathic effect in a monolayer culture of transplantable PT cells. Upon the onset of complete degeneration of the cells of the monolayer culture, the vessel with intestinal segments is removed from the roller, transferred to the box. The nutrient medium containing the virus is drained through the bottom side fitting. Discharged from the vessel Krsh1ki placed in a horizontal

The tweezers are used to remove intestinal segments from the holders, transfer them to a vessel and store them in a frozen state. Infected intestinal tissue is used as a starting material for the preparation of diagnostic β-antigens and other purposes.
The titers of viral diarrhea accumulation in intestinal tissue are determined by growth on monolayer PT cell culture. The accumulation of diarrhea virus in intestinal tissue by 1 ± 0.2 log. Exceeds the virus titer in monolayer cell culture.
Example 3: Cultivation of a coronavirus when growing intestinal tissue using the proposed method. Selection, preparation and planting of intestinal tissue on the holders is carried out as in example 1, then they proceed as in example 2, except that a coronavirus is taken instead of the diarrhea virus.
The control of the reproduction of the coronavirus was carried out with the help of DGA (gemagglutination reaction), which was made micro by the method in the plates with the wells of the Tokacchi apparatus. The titer of the coronavirus hemagglutinin in the infected tissue of the segments of the intestine exceeds the titer of the coronavirus in the starting material (suspension of feces from sick animals) by 8-10 hemagglutinating units, which indicates the accumulation of the virus biomass in the tissue. Specificity of coronavirus infection is exposed to HGS with a specific coronavirus antiserum.
П р и м е р 4. Cultivation of rotavirus in the cultivation of intestinal tissue by the proposed method. Selection, preparation and planting of intestinal tissue on the holders is carried out as in example 1, then as in example 2, only rotavirus is taken instead of diarrhea virus.
31451163Reproduction of rotavirus in the tissue of the segments of the intestine is controlled by RDP (diffusion precipitation reaction) with anti-tavirus serum. As the rotavirus antigen, they take the liquid portion of the suspension, pounded in the mortar of intestinal tissue. RDP is considered positive if there are clearly defined bands between the holes with the test material31451163
