Haemophilus influenzae. This invention relates to microbiology and relates to nutrient media for the isolation of Haemophilus influenzae.
The purpose of the invention is to increase the productivity of H. influenzae.
The use of the medium is illustrated by the following examples.
Example 1. 10.2 g of agar-agar for swelling was added to distilled water. The mixture is then aggravated until the agar is completely melted, then it is cooled to 40-50 ° C and the following ingredients are subsequently introduced1, g: casein-yeast hydrolyzate 40.8, glucose 0.6, iron nitrate 0.04, 2-3-indolindione 0.03. The pH of the medium is set at 7.410.2, filtered and poured into vials, autoclaved at 115 ° C for 30 minutes. The medium is cooled to 40-50 ° C, poured into Petri dishes (20-25 ml) or into test tubes {5 ml for canted agar).
0,035.Example 2. The medium of Example 1 is prepared, but the ingredients are used in the following ratios, g: agar-agar 10.8, casein-yeast hydrolyzate 41.8, glucose 0.62, iron nitra 0.043, 2- 3 indolindione 0.035.
0,04.Example 3. The medium of Example 1 is prepared, but the ingredients are used in the following ratios, g: agar-agar 11.0, casein-yeast hydrolyzate 44.0, glucose 0.65, ferric nitrate 0.045, 2-3- indolindione 0.04.

Выращивание ведут в термостате при 36 - в услови х повьш1енного содержани углекислого газа.for 15–20 min at 37 ° C. Growing up is carried out in a thermostat at 36 - under high carbon dioxide conditions.
Before sowing, cultures are washed off with saline and a number of dilutions are prepared. The seeding dose (0.1 mm) contains 1000 microbial cells (M. cells) and in 0.05 ml of the suspension 100 m. Cells.

. influenzae стрептококка и стафилококка возможна их дифференциаци .mixtures of N. cultures. influenzae streptococcus and staphylococcus their differentiation is possible.
H. influenzae bacteria form on the medium with smooth edges, smooth, tender, slightly grayish colonies with a diameter of 0.5 mm. Colonies of streptococcus are small, opaque, with a dense center. Staphylococcus colonies are large, up to 2 mm in diameter, smooth, with a smooth edge, the pigment is well pronounced.
Предлагаема среда суха , дл приготовлени ее используетс непищевое сырье и реактивы отечественного производства .The proposed medium is dry; non-edible raw materials and reagents of domestic production are used for its preparation.
1.When using a dry variant of the medium, a portion of 52-56 g is poured into the flask with 1000 ml of distilled water, left to swell for 5 minutes, then boiled over low heat for 2 to 3 minutes, filtered and then carried out in accordance with Example 1.
