Сульфокатионит может быть использован в ионообменной хроматографии аминокислот, пептидов , Сахаровj гидролизатов белков в научно-нрследовательских лаборатори х , й медицине, в сельском хоз йстве .This invention relates to the chemistry of high molecular weight compounds, and specifically to a method for producing a sulfonated microspheric styrene divivinyl benzene (DVB) copolymer (sulfonic cation exchanger). Sulfo cation exchanger can be used in ion exchange chromatography of amino acids, peptides, sugars protein hydrolysates in research laboratories, medicine, in agriculture.
The purpose of the invention is to improve the chromatographic properties of the supined product.
Example 1. In a 1 L jar of eMKdCTbro, 100 g of copolymer and 100 ml of DCE are placed, mixed, tightly capped and left to swell for 14 hours. A mixture containing 100 g of microspherical styrene-DVB copolymer (the amount of DVB in the copolymer is 8%), pre-swollen in 100 ml of dichloroethane (DCE) and 320 g of sulfuric acid (, 828), is placed in a 1-liter reactor. The mixture is stirred frame mixer. To prevent overheating, 360 g (50%) oleum is slowly introduced. Sulfonation proceeds at a ratio of reagents - copolymer: sulfuric acid: oleum 1: (3.23, 5): (3.6-4.0). The reaction temperature is 80 ° C. After 8 hours, stirring and heating of the reaction mass is stopped and the sulpholymass is poured into a container with 10 liters of cold distilled water to stand up from the cake particles. A day later, the undiluted granular copolymer is drained, and the precipitate is washed on the filter from excess sulfuric acid and dried. The static exchange capacity (SOY) for a 0.1 N solution of NaOH is 5.3 mg eq / g and is close to theoretical. The resulting cation exchanger is fractionated by a flotation method using a fractionation vessel, in which the walls of the conical part have an inclination to the axial line of less than 7. The chromatographic properties of the sorbent are tested under the conditions of the AAA-881 amino acid analyzer (Mikrotehn, Prague, Czechoslovakia). The data of chromatographic separation of hardly separated pairs of amino acids is given in the table.
40„As can be seen from the table, an increase in the time of copolymer sulfonation to 8.5 h leads to an improved separation of the threonine-serine pair. A sulfi40 „
The above 8 h results in improved separation of the threonine-serine pair to the detriment of the separation of the glycynalanine, tyrosine-phenylalanine pairs.
