Abstract

A method for producing lipids from blood cells by extracting chloroform: methanol: water with a single-phase solvent system, characterized in that, in order to increase the yield and shorten the time required to obtain the target product, sodium sulfate is added to the cell suspension at a concentration of 0.3-1, 0 M

Description

СПSP

оabout

This invention relates to biochemistry, in particular to lipid extraction methods.

The purpose of the invention is to increase the yield and reduce the time to obtain the target product.

Example. Donor blood stabilized with citrate solution of TsNIIGPK-76 was centrifuged at 1500 rpm in the cold for 20 minutes. The cell pellet was washed twice with physiological saline solution, suspended in cold 0.9% NaCl (1: 3 by volume), and centrifuged at 1500 rpm for 15 minutes.

Then, 2 ml of cell sediment, 300 mg of Na2SO4, 2 ml of NaO and 15 ml of chloroform: methanol (2: 1 by volume) were mixed. The mixture was centrifuged, the extract was taken, and the precipitate was re-extracted with chloroform: methanol: water (1: 2: 0.8 by volume) for an hour. The extract was diluted in a separatory funnel with water (4: 1 by volume) and after shaking and subsequent settling, the lower layer was taken, which was mixed with benzene (1: 1 by volume) and evaporated at 40 ° to obtain lipids in the amount of 96% of their total cell content.
