Известны способы ионного и ковалентного присоединени гепарина к полимерам С 13. Однако ионное присоединение способствует быстрому вымыванию гепари на с поверхности полимера, а в резу тате ковалентного св зывани гепари значительно дезактивируетс . Известен способ получени тромбо резистентных полимерных изделий, контактирунлцих с кровью, путем их обраЪотки поверхностно-активным ве: цecтвoм (ПАВ), например цетйламмоний-хлоридом , гепарином, глутаровым альдегидом. Этот способ не измен ет активност| гепарина 2 . Недостатками способа вл ютс применение в качестве ПАВ токсичных органических аминосоединений, обратима сорбци ПАВ на поверхности полимерных материалов, что отрицательно вли ет на прочность св зывани гепарина и снижает врем действи антикоагул нтного покрыти , при менение повышенной температуры (60-90°) и в р де случаев предварительной обработки поверхности материалов в окислительных средах. Цель изобретени - увеличение вр мени действи антикоагул нтного покрыти „ Цель достигаетс тем, что соглас но способу гепаринизации полимерных изделий, контактирукнцих с кровью, путем их обработки поверхностноактивным веществом (ПАВ), гепарином глутаровым альдегидом, в качестве ПАВ используют 1 - 2%-ный водный раствор, альбумина человека или 0,5%-ный физиологический раствор фи риногена и обработку ими провод т при 30-40°С 1,5-2,5 ч, отмывают изд лие дистиллированной водой, обработ ку гепарином производ т из водного раствора концентрации 40-5000 ед/мл при 30-40 С 1-1,5 ч, а глутаровым альдегидом - при 50-60 С 15-30 мин. Предлагаемым способом возможна обработка как пленок и катетеров, так и сосудистых протезов. Оптималь ными параметрами по количеству максимально необратимо-сортированного белка вл ютс : дл альбумина - 12%-ный водный раствор,а дл фибрине- . гена - 0,5 %-ный рчствор в физиологическом растворе, температура 30-40С и продолжительность 1-5-2,5 ч. При концентраци х белка выше указанных и продолжительности обработки больше 2,5 ч количество необратимо сорбированного белка на поверхности полимера не мен етс . Температура сорбции белка выше нежелательна из-за возможной денатурации белка. Оптимальными с точки зрени присоединени достаточного коли11ества гепарина вл ютс параметры: концентраци 40-100 ед/мл дл полимеров в виде сосудистых протезов, изготовленных из волокон фтррлона и лавсана,: температура 30-40С, продолжительность 1-1,5 ч. При концентрации 40 - 100 ед/мл на г ладкую полимерную поверхность присоедин етс 20-30 мкг/см гепарина, в результате чего достигаетс достаточный тромборезистентный эффект. Дл присоединени того же количества гепарина на поверхйость гофрированного тканного сосудистого протеза из волокон фторолон-лавсана из-за большой разветвленности поверхности необходима концентраци гепарина в растворе 3000-5000 ед/мл. Гистологические исследовани показали, что при такой обработке сосудистого протеза в отличие от обработки его раствором концентрацией гепарина 40-100 ед/мл, через 2-3 мес после имплантации в организм животного внутренн капсула протеза состоит из фиброзной ткани и воспалительна инфильтраци в ней практически отсутствует . Обработка глутаровым альдегидом проводитс дл уменьшени вымывани гепарина. Оптимальным режимом обработки глутаровым альдегидом вл етс : концентраци - 1 %, температура 5060°С , продолжительность обработки 15-30 мин. Пример 1.
