с Изобретение относитс к микробй логической промвдоленности и может быть использовано дл биологическо очистки сточных вод от органически соединений, в частности от фенольных и серосодержащих соединений, на предпри ти х целлюлозно-бумажно нефт ной, углехимической и других областей промышленности. Известен штамм Citrobacterfгеundii 21 дл очистки сточных вод от додецилсульфатов. Известен штамм Pseudomonas sp 1 дл очистки промышленных сточных вод от алкилсульфатов til. Известен также способ очистки сточных вод от фенолов Грибами С21 Недостатками этих штаммов вл етс невысока активность деструкции по отношению к фенольным соеди нени м.,Целью изобретени вл етс штам который способен утилизировать фено лыи лигнин. Поставленна цель достигаетс тем, что используют штамм черных дрожжей вида Exophiala nigrum R-11 который осуществл ет биологическую очистку от фенолов и лигнина. Культура ; черных дрожжей, иден тифицированных как Е nigrum J твапелена из проб байкальской воды, отобранных в прибрежной зоне Южног Байкала с использованием мембранны фильтров. Штамм R-11 вьщелен методом клон ровани с применением микрсмианипул тора и последующего рассева. Получ но около сорока клонов, послуживши материалом дл селекции и токсикогенетических испытаний. Штам14 черных дрожжей Exophiala nigrum R-11 хранитс в коллекции лаборатории генетики НИИ биологии Иркутского госуниверситета и в кол лекции ВНИИ генетики промышленных микроорганизмом (Москва), где ему присвоен регистрационный номер ЦМПМУ- 309 . После проверки на патогенность дл теплокровных животных и человека, а также дл злаковых сельскохоз йственных культур штамм R-11 рекомендован к внедрению в промыишенности, как непатогенный . Г1тамм черных дрожжей- Exophiala nigrum R-11 обладает следующими признаками. Морфологические признаки. В жидких средах при без встр хивани штамм образует овально-удлиненные клетки 5,2/8,3x1,9-4,7 мкм. Раз множение верхушечным или боковым почкованием, часто односторонним. Иi округлых крупных клеток образуют с гифы мицели . Мицелий 30-суточноП . культуры сильно развит, септированный , нерегул рно-разветвленный, в основном истинный, субстратный или воздушный. Культуральные признаки. При росте в жидкой глюкозо-пептонной среде или на сусле на 3 сут наблюдаетс помутнение, на дне осадок, не сна пленка, колько отсутствует или частичное . Через мес ц кольцо очень толстое, на поверхности среды обособленные дерновинки мицели . На дне незначительный осадок дрожжеподобных клеток, мути нет. На агаризованиых средах штрих пр мой, профиль выпуклый, край ровный , цвет черный блерт щий, консистенци Слабослизиста . Через мес ц штрих черный или буровато-серый, неравномерно-опущенный, поверхность складчата , край ворсинчато-окаймленный . Гигантска колони .достигает диаг метра 19-25 мм, выпукла , опущенна , буровато-сера с ровным крЬем, с приподн той центральной частью, окруженной гладким черным валиком, редкие радиальные складки. Физиологические признаки. Лэроб, образует черный пигмент, сахара не Сбраживает. Хорошо ассимилирует D .глюкозу, D-ксилозу, сахарозу, раство римый крахмал, глицерин, D-молочную кислоту, медленно ассимилирует ot-рамнозу, - -арабинсзу , малЬтозу, D-ijiaHHHT, D-сорбит, Слабо ассимилирует D-галактозу, рафинозу, этанол, дульцит, i-инозит, не ассимилирует, лактозу, лимонную кислоту, гексадекан . Из источников азота .ассимилирует нитриты, в среде без витаминов рост медленный, на среде с 50%-ной глюкозой не растет, выдерживает концентрацию Had, равную 7%. Оптимальна температура роста 20-25 С:, при 30-37С не растет. Чувствителен к 100 мкм/мл циклогексимида , обладает уреазной активностью, кислотообразование на меловом агаре отсутствует, желатин не разжижает, жир не гидролизует, эфир и крахмалоподобные соединени не образует. Шта№1 R-11 способен утилизировать фенолы. Пример 1. Штамм черных дрожжей Expplviala nigrum R-11 выращивают на сусло-агаре или. глюкозопептонной среде при , делают смыв, В 100 мл воды, содержащей пирокатехин или любой другой фенол: гидрохинон , фенол, гва кол в концентрации 10 м, ввод т суспензию 2 г черных дрожжей и через 24 ч определ ют остаточное содержание фенолов методом потенциометрического титровани . Степень очистки от пирокатехина 96,6%, от гидрохинона - 86,2%, от фенола - 04,3%, гва кола - 82,3%.
