Description
Spôsob prípravy sterolových alkoholov z lanolinuProcess for preparing sterol alcohols from lanolin
Oblasť technikyTechnical field
Vynález sa týka spôsobu prípravy sterolových alkoholov z lanolinu. Tieto sterolové alkoholy pozostávajú hlavne z cholesterolu a lanosterolov. The invention relates to a process for the preparation of sterol alcohols from lanolin. These sterol alcohols consist mainly of cholesterol and lanosterols. The mixture of these alcohols is used in the pharmaceutical industry for the synthesis of stereides and in cosmetics as an effective emulsifier. In particular, the starting material lanolin, which is obtained by washing wool, contains esters of higher fatty acids with sterol-type alcohols, in particular n-alcohols, optionally with diols. Alcohols bound in lanoline are referred to collectively as lanolin alcohols and the higher fatty acids bound in lanoline are referred to collectively as lanolin acids.
Doterajší stav technikyBACKGROUND OF THE INVENTION
According to prior art processes for the preparation of lanolin alcohols, lanolin is subjected to hydrolysis in an alkaline liquor and a lower alcohol at normal or elevated pressure. The hydrolysis produces a mixture of alkaline soaps, lanolinine acids and free lanolin alcohols, in particular cholesterol and lanosterols, in addition to the remainder of the esters, especially n-alcohols, which are difficult to hydrolyze. Extraction with a suitable solvent is used to isolate the lanolin alcohols from the mixture after hydrolysis, e.g. Pat. 2 619 495 and MS. of the author's certificate no. 231 912, using low solubility of alkaline soaps in the extracting agent. The disadvantages of these processes are the high consumption of solvents in the multiple extraction and the necessity to carry out the extraction at elevated temperatures around the boiling point of the extraction agent. It is also known to the procedure of US. pat. No. 88,805, in which the alkaline soaps of lanolinic acid are decomposed with a mineral acid and the free lanolinic acid is then converted into calcium or magnesium reindeer, which is insoluble in solvents, and subjected to extraction. Its disadvantage is that the non-hydrolyzed esters present and other hydrocarbon-type substances are present in solution together with the lanolin alcohols, contaminating, diluting and coloring them. Their presence also makes it difficult to separate calcium salts, as slime-like, hardly filterable suspensions are formed.
Podstata vynálezuSUMMARY OF THE INVENTION
This drawback is eliminated in the process according to the invention, which is based on the fact that a distillate obtained from a mixture of free lanolin alcohols, free lanolin alcohols, esters and other hydrocarbon-type distillation by means of a short-path evaporator with a wiped-off film is used to prepare the lanolinine acid calcium salts. at a temperature of 200 to 280 ° C and a non-condensable gas pressure of 10 1 to 10 2 Pa, the feed distillate by weight being 60 to 75% and its acid number 65 to 80 mg KOH / g.
The process of the invention has several advantages. By distillation of the mixture after hydrolysis and decomposition of the alkaline soaps, the more volatile lanolin alcohols and lanolin acids are separated in the distillate, while the non-hydrolyzed esters, especially the difficult decomposable lanolin acid esters with n-alcohols, remain in the distillation residue together with the bituminous and color products. On the other hand, these esters remain intact and chemically unchanged for possible isolation of other physiologically active ingredients. The acid value of the distillate after removal of the neutral distillation residue increases relative to the parent material, which is significant in terms of dividing the calcium soap suspension into solution. This phase separation is considerably better in the case of higher acid numbers, where hardly filterable gel structures are not formed. The lanolin alcohol concentrate prepared by the process of the invention is light yellow in color and does not require additional purification. Under the conditions of molecular distillation in a short path evaporator with gentle treatment of thermolabile substances, no decomposition reactions occur due to elevated temperatures. Since, under mild hydrolysis conditions, there is practically no hydrolysis of the lanolinic acid esters with n-alcohols and the water-soluble diols do not pass into the organic layer, the lanolin alcohols obtained at the end of the process are virtually pure sterol alcohols.
Molten lanolin is added to a solution of alkaline sodium hydroxide or potassium hydroxide ΚΟΉ in a mixture of a small amount of water with a lower alcohol such as ethanol or isopropanol and allowed to react for a sufficient period of time under stirring. The reaction mixture is then acidified with a dilute mineral acid such as hydrochloric HCl or sulfuric H2SO4, the precipitated organic layer is washed twice with water and the residual water and lower alcohol are removed therefrom, most preferably in a stripped evaporator under reduced pressure. The thus prepared mixture of free lanolin acids, free lanolin alcohols, esters and other organic substances is subjected to distillation on a short-pass evaporator with a film strip at a temperature of 200 to 280 ° C and a non-condensable gas pressure of 10 1 to 10 2 Pa, preferably on a multi-stage evaporator with increasing temperature in individual degrees. A distillate having a proportion of 60 to 75% by weight is obtained. with respect to the starting quantity for distillation. After distillation, the distillate is mixed with powdered calcium oxide in an amount corresponding to 1.5 to 2.5 times the equivalent amount according to the acid number of the distillate, and maintained at elevated temperature and stirring until calcium soaps are formed from the lanolinine acids present. After cooling, a suitable solvent such as acetone or gasoline is added to the mixture, the mixture is mixed and the resulting suspension is separated by settling or filtration. After removal of the solvent from the liquid phase, a mixture of sterol type lanolin alcohols is obtained.
Príklad uskutočnenia vynálezuDETAILED DESCRIPTION OF THE INVENTION
Po odstránení rozpúšťadla z filtrátu sa získa zmes cholesterolu a lanosterolov.40 g of NaOH are added in 1 liter of 90% ethanol in water and allowed to dissolve at elevated temperature with stirring. 400 g of molten lanolin are added to the solution and allowed to react for 5 hours. Hydrochloric acid is added in portions to the reactor in small excess relative to the hydrolysis liquor used. After vigorous stirring, the mixture was poured into a separatory funnel and washed twice with water at 75 ° C. The resulting organic phase was then passing the wiped film evaporator at 100 DEG C. and a pressure of 10 4 Pa freed from residual water and ethanol. The organic phase is a dark brown solid, solid at room temperature, with an acid number of 58 mg KOH / g. The mixture was subjected to distillation on a short-path, wiped-film evaporator at a heating oil temperature of 240 ° C and a non-condensable gas pressure of 12 Pa to give a distillate fraction of 72% and a distillation residue of 28% by weight of the starting material for distillation. The acid number of the distillate is 72 mg g / g, the distillate has a light yellow color. 250 g of distillate are mixed with 20 g of powdered calcium oxide, heated to a temperature of 90 to 120 ° C and allowed to react for 15 min with stirring. After cooling to 50 [deg.] C., 200 ml of acetone are added and, after thorough mixing, the warm suspension is filtered. After removal of the solvent from the filtrate, a mixture of cholesterol and lanosterols is obtained.
