FIELD: medicinal biotechnology, virology. SUBSTANCE: invention proposes the strain "Orloff-B", "ГКВ" N 2326 and describes the following strain properties: cultural-morphological characters (buoyant density is 1.18-1.20 g/$$$, titer = 4-6 lg $$$/0.5 ml and 7-8 lg $$$/0.5 ml on cells BHK-21); physiological-biochemical characters (optimum pH = 7.4, inactivation at 56 C for 30 min, sensitivity to ether and acetone); antigenic properties (hemagglutinating activity in vitro - 2-8 "ГАЕ"); immunogenic properties (100% formation of specific antibody in rabbits and guinea pigs, at reverse titer in "РТГА" - from 640-2560 to 1280-10240, respectively; serum conversion is 80-100%); virulent properties (strain does not show virulent or contagious properties); toxicity (no toxicity at dose 1000 $$$/0.5 ml in guinea pigs and mice). EFFECT: enhanced immunogenicity of vaccine against measles, improved properties of strain-producer. 3 tbl

Изобретение относится к медицинской биотехнологии. С целью повышения иммуногенности вакцины против краснухи в качестве продуцента предложен штамм "Орлов-В", ГКВ N 2326. Культурально-морфологические признаки: однородные сферические частицы с диаметром 50 - 70 нм; плавучая плотность в градиенте сахарозы 1,18 - 1,20 г/см; размножается с развитием цитодеструктивного действия до титра 4 - 6 lg ТЦД50/0,5 мл в клеточных культурах животных и человека; слабо размножается в первичных тканевых культурах куриных и перепелиных фибробластов; при роллерном или суспензионном способах культивирования на клетках ВНК-21 выход вирусной биомассы 7 - 8 lg ТЦД50/0,5 мл; не размножается при температуре ниже 20oС и выше 40oС. Физиолого-биохимические признаки: оптимум рН = 7,4; инактивируется в течение 30 мин при 56oС; чувствителен к эфиру и ацетону. Антигенные свойства: гемагглютинирующая активность in vitro 2 - 8 ГАЕ; агглютинирует эритроциты голубей, однодневных цыплят, человека с группой крови 0. Иммуногенные свойства: специфические антитела при контроле на кроликах и морских свинках образуются в 100% случаев и характеризуются обратным титром антител в РТГА от 640 - 2560 до 1280 - 10240 соответственно; наблюдаемость сероконверсии 80 - 100%. Вирулентные свойства: штамм не обладает вирулентными и контагиозными свойствами. Токсичность: штамм не токсичен при введении морским свинкам и мышам в дозе 1000 ТЦД50/0,5 мл (1 прививочная доза человека).The invention relates to medical biotechnology. Immunogenic properties: specific antibodies, when tested on rabbits and guinea pigs, are formed in 100% of cases and are characterized by an inverse antibody titer in rtga from 640 - 2560 to 1280 - 10240, respectively; the observability of seroconversion is 80 - 100%. Virulent properties: the strain does not have virulent and contagious properties. Toxicity: the strain is not toxic when administered to guinea pigs and mice at a dose of 1000 TCD 50 / 0.5 ml (1 human inoculation dose).
