Abstract

FIELD: biotechnology, immunology. SUBSTANCE: strain of hybrid cells is obtained by fusion of murine myeloma cells P3-X63-Ag8-653 with cells of peripheral lymphonodus from mice of strain BALB/c immunized two-fold (100 mcg) with IgM isolated from pig blood serum by chromatography methods. Proposed strain is a producer of monoclonal antibodies specifically reacting with pig IgM only. Monoclonal antibody preparations obtained from ascitic fluid can be used for identification and quantitation of pig IgM level by method of radial immunodiffusion and in immunoenzyme analysis. EFFECT: preparing the strain of hybrid cells indicated above.

Claims (1)

Цель: получение штамма гибридных клеток, продуцирующих моноклональные антитела к иммноглобулину класса М свиньи. В результате проведенной работы был получен штамм гибридных клеток путем слияния клеток мышиной миеломы линии P3-X63-Ag8-653 с клетками периферических лимфатических узлов (паховых и подколенных) мышей линии BALB/с, иммунизированных 2-х кратно (100 мкг) IgM, выделенным из сыворотки крови свиньи хроматографическими методами. Предложенный штамм является продуцентом моноклональных антител специфически реагирующих только с IgM свиньи, и не дающих перекрестных реакций с иммуноглобулинами других классов. Препараты моноклональных антител, полученные из асцитической жидкости, можно использовать для идентификации и количественного определения уровня IgM свиньи методом радиальной иммунодиффузии (РИД) и в иммуноферментном анализе (ИФА). Штамм найдет применение в научно-исследовательских институтах, проблемных лабораториях, ведущих работы в области иммунологии и биотехнологии.Purpose: to obtain a strain of hybrid cells producing monoclonal antibodies to pig immunoglobulin class M As a result of this work, a strain of hybrid cells was obtained by fusion of mouse P3-X63-Ag8-653 mouse myeloma cells with cells of peripheral lymph nodes (inguinal and popliteal) BALB / c mice immunized with 2-fold (100 μg) IgM isolated from pig blood serum by chromatographic methods. The strain will find application in research institutes, problem laboratories, leading work in the field of immunology and biotechnology.
