Abstract

FIELD: biotechnology. SUBSTANCE: strain of hybrid cells is obtained by fusion of murine myeloma cells Sp 2/0 with murine lymphocytes BALB/c immunized four-fold (200 mcg) with sIgA isolated from pig broncho-alveolar lavage by chromatography methods. Proposed strain is a producer of monoclonal antibodies reacting with L-chains of pig immunoglobulins only. Monoclonal antibody preparations obtained from ascitic fluid can be used for identification and quantitation of pig immunoglobulins level, for species blood proteins assay and study of pig L-chain immunoglobulins antigenic structure. Strain can be used in biotechnology and immunology. EFFECT: preparing the strain of hybrid cells indicated above.

Цель исследований - получение штамма гибридных клеток, продуцирующих моноклональные антитела к L-цепям иммуноглобулинов свиньи. В результате проведенной работы был получен штамм гибридных клеток путем слияния клеток мышиной миеломы линии Sp 2/0 с лимфоцитами селезенки мышей линии BALB/с, иммунизированных четырехкратно (200 мкг) sIgA, выделенным из бронхо-альвеолярных смывов свиньи хроматографическими методами. Предложенный штамм является продуцентом моноклональных антител, специфически реагирующих только с L-цепями иммуноглобулинов свиньи и не дающих перекрестных реакций с иммуноглобулинами других видов животных. Препараты моноклональных антител, полученные из асцитической жидкости, можно использовать для идентификации и количественного определения уровня иммуноглобулинов свиньи, для определения видовой принадлежности белков крови и изучения антигенной структуры L-цепей иммуноглобулинов свиньи методами ИФА, дот-блот ИФА и РИД. Предложенный штамм был апробирован с положительным результатом в лабораторных условиях в 1994 году. Штамм найдет применение в научно-исследовательских институтах, проблемных лабораториях, ведущих работы в области иммунологии и биотехнологии.The aim of the study was to obtain a strain of hybrid cells producing monoclonal antibodies to the L-chains of pig immunoglobulins. As a result of the work, a strain of hybrid cells was obtained by fusion of mouse Sp 2/0 mouse myeloma cells with spleen lymphocytes of BALB / c mice immunized four times (200 μg) sIgA isolated from pig broncho-alveolar swabs by chromatographic methods. The proposed strain is a producer of monoclonal antibodies that specifically react only with L-chains of pig immunoglobulins and do not cross-react with immunoglobulins of other animal species. Monoclonal antibody preparations obtained from ascitic fluid can be used to identify and quantify the level of pig immunoglobulins, to determine the species affiliation of blood proteins and to study the antigenic structure of pig immunoglobulin L chains by ELISA, ELISA dot blot and ELISA. The proposed strain was tested with a positive result in laboratory conditions in 1994. The strain will find application in research institutes, problem laboratories, leading work in the field of immunology and biotechnology.
