Изобретение относится к области физико-химического разделения веществ, а именно к матрицам для хроматографии и способам их получения и применения для очистки биологически активных веществ. Предложен способ получения микросферической гранулированной целлюлозы. Диспергирование проводят в трансформаторном масле, а выделение продукта осуществляют обработкой полученной смеси гексаном и ацетоном. На основе микросферической целлюлозы получают сорбент общей формулы (C2H5) -CH2-O-R, где R - матрица целлюлозы. Указанные сорбенты используют доя очистки стрептокиназы хроматографическим методом. Сорбцию проводят на диэтиламиноцеллюлозе в OH-форме в качестве сорбента, причем сорбент первоначально уравновешивают фосфатным буфером с электропроводностью 5,9±0,3 мксим/см, а затем элюируют фосфатным буфером с электропроводностью 10,0±0,6 мксим/см, причем весь процесс ведут при рН 7,0±0,1. В результате использования изобретения упрощается технология получения сорбентов на основе микросферической целлюлозы. Использование заявляемого сорбента позволяет существенно повысить выход стрептокиназы.The invention relates to the field of physico-chemical separation of substances, namely, matrices for chromatography and methods for their preparation and use for the purification of biologically active substances. Based on microspherical cellulose, a sorbent of the general formula (C 2 H 5 ) -CH 2 -OR is prepared, where R is the cellulose matrix. These sorbents are used for purification of streptokinase by chromatographic method. Sorption is carried out on diethylaminocellulose in the OH form as a sorbent, the sorbent being initially balanced with a phosphate buffer with a conductivity of 5.9 ± 0.3 µsim / cm, and then eluted with a phosphate buffer with a conductivity of 10.0 ± 0.6 µsim / cm, the whole process is carried out at pH 7.0 ± 0.1. As a result of using the invention, the technology for producing sorbents based on microspherical cellulose is simplified. The use of the inventive sorbent can significantly increase the yield of streptokinase.
