Способ относится к медицинской биотехнологии, биологической промышленности и ветеринарии, в частности к усовершенствованной технологии получения ферментативного гидролизата, а также к новой питательной середе на его основе для ультивирования клеток эукариотов - продуцентов биологически активных вещест, диагностических и профилактических биологических и медицинских препаратов. Сущность изобретения заключается в том, что измельченную мышечную ткань ластоногих, смешанную с дистиллированной водой в массовом соотношении 1 : 2, подвергают гидролизу в присутствии ферментсодержащих органов ластоногих, взятых в количестве 15 об.%, при 40 - 42oС в щелочной зоне рН 7,6 - 7,8 в течение 4 - 5 суток в присутствии хлороформа с последующим прогреванием биомассы при 80 - 90oС в течение 5 - 10 мин, осаждением высокомолекулярных соединений сначала в присутствии 20 - 22%-го раствора хлористого кальция, затем - в изоэлектрической точке белка, фильтрованием препарата и высушиванием. Сущность изобретения заключается также в том, что питательная среда "Целат" для культивирования клеток эукариотов содержит ферментативный гидролизат мышечной ткани ластоногих, сыворотку крови крупного рогатого скота и раствор Хэнкса при следующем соотношении компонентов, об.%: ферментативный гидролизат мышечной ткани ластоногих - 0,15 - 0,20; сыворотка крови крупного рогатого скота - 5 - 10; раствор Хэнкса - остальное. Предлагаемый способ получения ферментативного гидролизата позволяет упростить технологию процесса и снизить себестоимость конечного продукта за счет использования отходов зверобойного промысла - мышечной ткани ластоногих, а в качестве фермента - неутилизируемых отходов того же промысла - ферментсодержащих органов ластоногих. Питательная среда "Целат" на основе ферментативного гидролизата мышечной ткани ластоногих обладает высокими ростстимулирующими свойствами для некоторых первичных, а также перевиваемых гетероплоидных клеток животных и человека, что позволяет применять ее для получения больших количеств клеточной биомассы в биотехнологических производствах вместо многокомпонентной синтетической питательной среды 199, аминокислота для получения которой закупается за рубежом.The method relates to medical biotechnology, biological industry and veterinary medicine, in particular, to an improved technology for producing an enzymatic hydrolyzate, as well as to a new nutrient medium based on it for the efficacy of eukaryotic cells producing biological active substances, diagnostic and prophylactic biological and medical preparations. The invention also consists in the fact that the nutrient medium "Celat" for the cultivation of eukaryotic cells contains enzymatic hydrolyzate of muscle tissue of pinnipeds, blood serum of cattle and Hanks solution in the following ratio of components, vol.%: Enzymatic hydrolyzate of muscle tissue of pinnipeds - 0.15 - 0.20; blood serum of cattle - 5 - 10; Hanks solution - the rest. The proposed method of obtaining enzymatic hydrolyzate allows you to simplify the process technology and reduce the cost of the final product through the use of waste of the hunting industry - muslin tissue of pinnipeds, and as an enzyme - non-utilizable wastes of the same industry - enzyme-containing organs of pinnipeds. Nutrient medium "Celat" based on enzymatic hydrolyzate of muscle tissue of pinnipeds has high growth-stimulating properties for some primary as well as transplantable heteroploid cells of animals and humans, which allows it to be used to obtain large quantities of cellular biomass in biotechnological production instead of multicomponent synthetic nutrient medium 199, amino acid for the receipt of which is purchased abroad.
