Все методики для экспериментов на животной модели 3xFAD были одобрены Институциональным комитетом по уходу и применению животных (IACUC) в Ханьянском колледже медицины (Hanyang College of Medicine) под номером одобрения 2018-0047А. These experiments were performed according to NIH (National Institutes of Health) guidelines. To minimize bias, behavioral assays were primarily analyzed by two experimenters in a blinded fashion. Transgenic mice with Alzheimer's disease (3xTg-AD) (Jackson Laboratory, Maine, USA) at the age of 18 and 15 months were used in the experiments.

A needle (26 gauge) was left at the injection site for 5-10 minutes after completion of each injection and slowly removed. If inaccurate injection into the hippocampal and cerebral ventricle (ICV) sites was confirmed, these mice were excluded from analysis.

That is, the expression of Nurr1 and Foxa2 in brain cells has been identified to have a clinical gene therapy effect on Alzheimer's disease (FIGS. 5, 6, 7, 8 and 9).

Two months after administration, Alzheimer's disease transgenic mice were sacrificed, fixed in paraformaldehyde (PFA) and then blocked for 1 hour with 0.6% Triton X-100 in 1% BSA/PBS. Primary antibodies (p-tau, Tuj1) were then bound to the same solution and the tissue was stained overnight at 4°C. Secondary staining was performed with Cy3 (1:200, Jackson Immunoresearch Laboratories), Alexa Fluor 488 (1:200, Life Technologies) as secondary antibodies for imaging. Stained cells were mounted with VECTASHIELD, DAPI mounting solution (Vector Laboratories), and the entorhinal cortex region was imaged using a confocal microscope (Leica PCS SP5). The resulting images are shown in FIG. 10, and their quantitatively measured results are shown in FIG. 11 and Table 1.
