Abstract
Description
The invention relates to medicine, namely to medicines made in the form of suppositories for topical use - treatment of rectal injuries, cracks, inflammation.
Б. 2000 A61K 9/02, 35/78, А61Р 43/00) [1, 2].A number of products are known for topical use - a suppository for the treatment of rectal cracks and injuries (patent RU 15547 U1 IPC A61K 9/02 (2000.01), patent RU 2157185 C1 Suppositories for the treatment of proctological diseases Dozorov A.V., Pakhomov A.B. 2000 A61K 9/02, 35/78, A61R 43/00) [1, 2].
The disadvantage of these suppositories is their narrow spectrum of therapeutic action, mainly antimicrobial. They have limitations for use due to their individual intolerance, as well as the risk of developing side effects with long-term use - decreased sensitivity of microflora, metabolic disorders.
The closest analogue is a suppository for the treatment of rectal fissures (RU 2207138 C1 Rectal suppositories with propolis Ionova S.A., Novoselova E.A., Polunina E.E. 2002 A61K 35/64, 9/02) [3], containing propolis, dimexide, distilled monoglycerides, witepsol in certain ratios. The invention ensures the rapid onset of a pharmaceutical effect due to the rapid entry of the substance into the blood, while the proposed suppositories have high quality characteristics and broad pharmacological properties.
However, bee propolis used as one of the main ingredients is contraindicated for allergy sufferers; in addition, the suppository does not stimulate the body's immunocompetent systems, does not inhibit free radical reactions in the wound, which causes a slow wound healing effect, relapses of the disease and concomitant allergic reactions. Suppositories contain various oils as the main component. Our suppository does not contain a fatty component and is completely absorbed when administered rectally.
The task was set to create a highly effective wound-healing anti-inflammatory, immunomodulating, antibacterial and local anesthetic suppository, which has a pronounced clinical effect and has broad functionality.
%:The problem was solved by introducing a suppository containing a 50% aqueous solution of dimethyl sulfoxide, a 10% solution of hyaluronic acid, hydrolyzed collagen - 30%, Vitamin C (ascorbic acid) - 4.26%, Coenzyme Q10 (ubiquinone) - 2% . The suppository has the following ratio of components, wt. %:
Hyaluronic acid - 10 g.
Hydrolyzed collagen - 30 gr.
Vitamin C (ascorbic acid) - 4.26 g.
Coenzyme Q10 (ubiquinone) - 2 gr.
Dimethyl sulfoxide 50% aqueous solution - 100 g.
The parameters of the components introduced into the composition of the proposed suppository were determined experimentally. The introduction of components below the lower or above the upper limit does not provide an optimal therapeutic effect, and also worsens the presentation.
Characteristics of the components of the suppository.
Витамин С (аскорбиновая кислота) и коэнзим Q10 (убихинон) обеспечивает улучшение тканевого обмена, стимулирует метаболические процессы, коллаген гидролизованный стимулирует модулирующие процессы иммунитета (иммуномодулирующее действие).A 50% solution of dimethyl sulfoxide provides an antibacterial effect and an analgesic effect, hyaluronic acid is a suppository-forming component and has an anti-inflammatory, adaptogenic and stimulating effect on the enzymatic activity of tissues. Vitamin C (ascorbic acid) and coenzyme Q10 (ubiquinone) improve tissue metabolism, stimulate metabolic processes, hydrolyzed collagen stimulates modulating processes of immunity (immunomodulatory effect).
A comparative analysis with the prototype showed that the claimed composition of the suppository differs from the known one by the introduction of new components, namely a 50% aqueous solution of dimethyl sulfoxide, a 10% solution of hyaluronic acid, hydrolyzed collagen - 30%, vitamin C (ascorbic acid) - 4.26% Coenzyme Q10 (ubiquinone) - 2% in the new component ratio.
An analysis of known suppositories similar to the claimed suppository in terms of the spectrum of therapeutic action showed that the composition: 10% solution of hyaluronic acid, hydrolyzed collagen - 30%, vitamin C (ascorbic acid) - 4.26%, coenzyme Q10 (ubiquinone) - 2 %, a 50% aqueous solution of dimethyl sulfoxide, in which all of the indicated ingredients are soluble or compatible with dimethyl sulfoxide, has not previously been used in the practice of treating trophic ulcers and superficial wounds.
The suppository is prepared as follows.
The calculation is based on 100 kg of product. For example, 100 kg of a 50% aqueous solution of dimethyl sulfoxide is added to it with hyaluronic acid (10 kg), hydrolyzed collagen (30 kg), vitamin C (ascorbic acid) (4.26 kg), Coenzyme Q10 (ubiquinone) (2 kg). The resulting mass is thoroughly mixed for 45 minutes. Subsequently stored at a temperature of 2 to 5 degrees.
When prepared under the influence of stirring, a ratio of medicinal substances is created that allows one to achieve the desired wound-healing anti-inflammatory, immunomodulatory, antibacterial and local anesthetic effect. The proposed technical solution makes it possible to obtain an inexpensive, cost-effective wound healing agent with high efficiency against a wide range of diseases accompanied by the formation of rectal fissures, as well as to reduce treatment time.
The inventive suppository is very plastic, easy to administer rectally, has high penetrating ability at the injection site, is non-toxic, and also has antibacterial and antifungal activity due to its constituent components (dimethyl sulfoxide) [11, 12, 13]. Hyaluronic acid creates a protective layer on the wound surface that promotes healing. The suppository stimulates the body's immunocompetent forces, is harmless and has no side effects. Due to the included components and their ratio, a technical result was obtained - a wound-healing, anti-inflammatory, immunomodulating, antibacterial and local anesthetic effect of the suppository.
