Prior Art
Antlers (Cervi Parvum Cornu) refer to the unossified young antlers of deer and, along with ginseng, have long been recognized as the most representative tonic material and are therefore widely used in Korea, China and Japan. The pharmacological property of a natural medicine with such a tonic effect is characterized by complex effectiveness, manifested throughout the body, and not by selective manifestation of activity and effect on individual organs. The pharmacological activity of antlers includes stimulation of the synthesis of proteins and nucleic acids, stimulation of hematopoiesis, strengthening of the immune system, effects on the cardiovascular system, anti-stress effects, anti-aging effects and anti-ulcer activity. Gangliosides, pantocrine, amino acids, calcium phosphate, calcium carbonate, collagen, phospholipids, chondroitin, glucosamine, hyaluronic acid, etc. are known as pharmacologically active components of antlers.
Sialic acid, known as the activity marker material of antlers, is a monosaccharide that attaches in the form of a sugar chain to a cell or soluble protein. It is a constitutional material that plays an important role in neuronal transmission, the formation of ganglion cells in the brain, and the maintenance of ganglioside structure and function. The most common example of sialic acid is N-acetylneuraminic acid.
Glycosaminoglycans (GAGs) are mucopolysaccharides having a skeleton in which uronic acid and glucosamine are repeated with partial sulfation. GAGs exist in the body in the form of proteoglycans. S-GAGs are the main component of cartilage in joints, nucleus pulposus of discs, etc., and are known to have therapeutic effects on arthritis, potent anti-inflammatory effects, and water-retaining effects.
Bacillus coagulans is a Gram-positive facultative anaerobic bacterium that grows in the temperature range of 30-55°C, with an optimal temperature for growth of 50°C. It exhibits characteristics of both Bacillus and Lactobacillus genera, exhibiting spore formation and lactic acid production. Bacillus coagulans has attracted attention as a strain that can produce L-lactic acid, heat-stable enzymes, and the antimicrobial peptide coagulin. The ability to produce biomaterials at high temperatures also has the advantage of minimizing contamination problems.
Various studies on antlers are currently being conducted. There are reports of many studies aimed at extraction using water-soluble and organic solvent (alcohol, chloroform, ethanol, etc.), extraction of useful components from fermented antlers using protein hydrolytic enzymes and microorganisms, as well as analysis and research of various markers materials present in antlers.
Korean patent KR 1776686 describes a method for preparing a solid preparation from a fermented antler product, and Korean patent KR 1336292 describes a method for extracting effective components from antlers using an ultra-high pressure, low temperature extraction process. However, these methods differ from the method of the present invention, in which the extract of fermented antlers is obtained by three-step processing.

Ltd., England). To 10 g of the test sample, 5% TCA (trichloroacetic acid) solution was added in the same amount as the test sample, and the mixture was stirred for 1 hour. The mixture was then centrifuged at 10,000 rpm for 10 minutes to remove proteins, and the supernatant was collected. 100 ml of diethyl ether was added to the supernatant and then extraction was carried out in a repeating mode, i.e. three times to remove lipids, pigments and other lipophilic materials. The resulting aqueous layer was concentrated under reduced pressure at 40°C and dissolved in 10 ml of 0.2 N lithium citrate buffer (pH 2.2). It was then filtered through a membrane filter (pore size 0.2 μm, Advantec MFS, Japan) and analyzed on an automatic amino acid analyzer (Biochem 20, Pharmacia Biotech. Ltd., England). 1. The following Table 1 shows the results of comparison of the physicochemical characteristics of the fermented antlers extract prepared in accordance with the high temperature and high pressure processing conditions in step (1) to obtain the fermented antlers extract in Preparation Example 1.
Physico-chemical characteristics of fermented antlers extract depending on various processing conditions at high temperature and high pressure
There is a tendency for pH to decrease as pressure and processing time increase. This may be due to changes in the physical properties of the raw material when processed at high temperature and high pressure, which leads to greater recovery of acidic components among the effective components recovered. Sweetness and soluble solids content also increased at higher pressure and longer processing time. In terms of color, L represents brightness, a represents redness, and b represents yellowness. Redness tends to increase with higher pressure and duration of treatment.
Example 2. Content of effective components in the extract of fermented antlers depending on various processing conditions at high temperature and high pressure
(1).The following Table 2 shows the results of comparison of the content of sulfated glycosaminoglycans (s-GAG) and N-acetylneuraminic acid between the fermented antlers extract of Preparation Example 1 and the fermented antlers extract obtained in accordance with the method of Preparation Example 1, but subject to processing at high temperature and high pressure in stage (1).
Glycosaminoglycans are mucopolysaccharides composed of repeating structures of amino sugars and uronic acid or galactose, and they are known to be essential components in animal tissues and connective tissues. In particular, sulfated glycosaminoglycans play a role as a major component of cartilage in joints and help maintain joint moisture, thereby being effective in treating diseases such as arthritis. They form proteoglycans by binding to proteins and are present in various connective tissues such as skin and umbilical cord, regulating their structure and function to maintain the integrity of each tissue.
N-Acetylneuraminic acid is a monosaccharide and is found in cell glycoproteins, cell membranes and mucus, as well as glycolipids such as ganglioside, as a major constitutional component of the membrane of the brain and nerve cells. N-Acetylneuraminic acid is a representative compound of sialic acid.

Table 5 below presents the results of a comparison of the free amino acid content of fermented antlers extract depending on various processing conditions.
Table 5. Content of effective components in the extract of fermented antlers depending on various processing conditions
1. As a result, the fermented antler extract from Comparative Example 2 showed the lowest content, while the fermented antler extract from Preparation Example 1 showed the highest content. The total essential amino acids (threonine, valine, methionine, isoleucine, leucine, phenylalanine, tryptophan and lysine) also showed the highest value in the fermented antlers extract from Preparation Example 1.
Essential amino acids cannot be synthesized in the human body and must be obtained from food or supplements. Among the essential amino acids, leucine plays a role in stimulating protein synthesis, promoting the growth of muscle tissue and strengthening skeletal muscles, etc. Lysine is involved in calcium metabolism, helping to maintain calcium absorption and nitrogen balance, thereby promoting bone growth and development. When consumed together with vitamin C, lysine promotes the synthesis of collagen, which is a necessary component for maintaining the elasticity of the skin and blood vessels, and the formation of cartilage. Phenylalanine is an essential substance for the production of neurotransmitters such as tyrosine, dopamine, epinephrine and norepinephrine. It is known to help with brain cell regeneration and protein synthesis. The fermented antler extract from Preparation Example 1 exhibits high levels of these amino acids, suggesting that it may promote body growth and brain development.
Example 5. Solubility of fermented antlers extract in water
In fig. 1 shows the water solubility level of the fermented antlers extract powder from Preparation Example 1. Using the fermented antlers extract powder obtained by pre-treatment at high temperature and high pressure, the solubility level of the powder was determined at 30 second intervals, i.e. immediately after adding to 1 minute and 30 seconds after adding. The results showed that the powder began to dissolve slightly immediately after addition, and that approximately half the volume of the powder had dissolved within 30 seconds of addition. Most of the powder dissolved after about 1 minute, and in the last 1 minute and 30 seconds complete dissolution occurred, causing the layers to separate due to the lipid components of the antler extract. Thus, it should be recognized that due to the rapid solubility in water of fermented antlers extract powder pre-treated at high temperature and high pressure, it can be used in various products and formulations, and at the same time useful components with low molecular weight obtained as a result of processing at high pressure, will be easily absorbed in the human body.
Example 6. Storage stability of fermented antlers extract
The following Table 6 shows the results of a storage stability study of the fermented antler extract powder obtained in Preparation Example 1. After various storage periods, the total bacterial count remained negative or 1 CFU/g throughout the storage period, and the entire Escherichia coli count was negative. However, the pH values ranged from 5.71 to 5.78 during the storage period, and thus the powdered product is considered to remain stable throughout the storage period.
