Blots were visualized using Pierce™ ECL Substrate (ThermoFisher Scientific, Cat. #32106) and chemiluminescence was monitored using a DIANA III Imaging System or a ChemiDoc™ MP Imaging System (BioRad). To analyze the effect of the compound on the amount of PAH protein, densitometric analysis was performed using ImageJ software.

This measurement determines changes in the heat generated or absorbed when molecules interact, generating binding kinetics and a thermodynamic profile of the molecular interaction.

Twelve 3 μl injections of the compound/ligand mixture were titrated into the sample cell (containing the protein) over 31 minutes at a stirring speed of 750 rpm. The data were adjusted to the baseline values by subtracting the baseline data obtained from equal injections of the compound into the buffer solution. Titration curves were analyzed using the analytical software MicroCal PEAQ-ITC (version 1.1.0.1262, Malvern Instruments Ltd.) using a model that takes a set of identical binding sites, where the total heat of the solution is calculated as follows:
