Изобретение относится к биотехнологии и медицине, в частности к получению нуклеозида 3-бутилтио-1-(бета-D-2-дезоксирибофуранозил)-5-фенил-(4Н)-1,2,4-триазола (соединение (1), фиг. 1) (БДФТ) и обнаружению его противовирусного действия. Изобретение может быть использовано для лечения и профилактики герпетической инфекции человека, в научно-исследовательских лабораториях, химико-фармацевтической промышленности, лечебных учреждениях.The invention relates to biotechnology and medicine, in particular to the production of 3-butylthio-1-(beta-D-2-deoxyribofuranosyl)-5-phenyl-(4H)-1,2,4-triazole nucleoside (compound (1), Fig. 1) (BDFT) and the detection of its antiviral effect. The invention can be used for the treatment and prevention of human herpes infection in research laboratories, the chemical and pharmaceutical industry, and medical institutions.
Модифицированные нуклеозиды, по своей природе являющиеся антиметаболитами, проявляют широкий спектр антивирусной активности. В современной лечебной практике активно используются препараты Ацикловир, Видарабин, Азидотимидин, и Рибавирин в комплексной терапии герпетических инфекций, ВИЧ и гепатита С, соответственно [Looking Back in 2009 at the Dawning of Antiviral Therapy Now 50 Years Ago: An Historical Perspective. Отдельного внимания заслуживают нуклеозиды 1,2,4-триазола, проявляющие антивирусную активность широкого спектра против гриппа А и В, респераторно-синтициальной вирусной инфекции, цитомегаловируса (ЦМВ), вируса герпеса простого 1 и 2 типов, вируса герпеса 6 типа. Для определения антигерпесвирусной активности использовали эталонный штамм L2 вируса герпеса простого типа 1 ( ВПГ-1/ L 2 ), полученный из Государственной коллекции вирусов Института вирусологии им. Д.И.Ивановского (ФГНИЦЭМ им. Н.Ф. Гамалеи МЗ РФ). После 48 часов инкубации, когда в контроле вируса развивался 95-100% цитопатический эффект (ЦПЭ), определяли концентрации, ингибирующие развитие вирусиндуцированного цитопатического эффекта по сравнению с контролем на 50% (ИД50) и практически полностью (ИД95).The antiviral activity of compound (1) was studied in vitro in Vero E6 cell culture.
