Б. 2021. - №8. - С. 141-144).The process of lipid peroxidation (LPO) is commonly understood as the chemical process of lipid oxidation under the influence of free radicals. LPO reactions can lead to disruption of the structure of all cell membranes and cell damage, which is a mechanism of pathogenesis of a number of diseases. At the same time, LPO is physiological, providing regulation of the structure and implementation of the functions of membranes. In this connection, maintaining a balance between these processes ensures the physiological course of many processes (Lipid peroxidation: “harm” and “benefit” / Lebedev I.A., Burundukova P.I., Abdullina M.B. // Notes of a scientist. 2021. - No. 8. - pp. 141-144).
Н.Э. 2021. - Т. 247. - №3. - С. 80-85).Initially, the formation of diene conjugates of fatty acids (DCFA) occurs. With further exposure to hydroxyl radicals, lipid hydroperoxides are formed. The resulting peroxidation products affect the phospholipid components of membranes and cell structures, disrupting the functioning of organelles and the body as a whole. Under the influence of free radicals, fatty acids decompose into final oxidation products: aldehydes, peroxides, diene conjugates (DC), malondialdehydes (MDA), Schiff bases (The influence of antioxidant drugs of different mechanisms of action on the level of lipid peroxidation products in the body of pregnant ewes / Kashirin L.G., Dorokhina Yu.E., Trfandyan M.T. // Scientific notes of the Kazan State Academy of Veterinary Medicine named after N.E. Bauman. 2021. - T. 247. - No. 3. - P. 80-85 ).
Antioxidants are natural or synthetic substances that reduce or neutralize the oxidative effect of free radicals (Features of lipid peroxidation processes in the body of cats with urinary tract diseases and their correction with commercial feeds / Vorontsova O.A., Pudovkin N.A., Salautin V.V. ., Klyukin S.D. // Agrarian scientific journal. 2021. - No. 4. - pp. 37-40). Substances with antioxidant capacity include:
а) ферменты - супероксиддисмутаза, глутатионпероксидаза и другие;a) enzymes - superoxide dismutase, glutathione peroxidase and others;
б) вещества пептидной природы - глутатион и другие;b) substances of peptide nature - glutathione and others;
в) экзогенные антирадикальные вещества - ретинол, аскорбиновая кислота, токоферол и глутатион и другие;c) exogenous antiradical substances - retinol, ascorbic acid, tocopherol and glutathione and others;
2021. Т. 24. №4. С. 51-54).d) synthetic antiradical substances - meldonium, probucol, pentahydroxyethylnaphthoquinone, methyl-ethylpyridinol, butylated hydroxytoluene and others (Providing the body with vitamin D in therapeutic and dental patients / Boldyreva Yu.V., Kurlovich N.A., Lebedev I.A., Bragin A.V., Shikhshabekova U.A., Zaitkhanova K.E. // Questions of biological, medical and pharmaceutical chemistry. 2021. T. 24. No. 4. pp. 51-54).
А., Гумеров A.M. Fullerene C60 DerivativesasEfficientSensitizersofOxidationundertheMildConditionsofAtmosphericAir // Russ. J. of Gen. Chem. 2017. - Vol. - 87. - №10. - P. 2497-2499). C60 fullerene derivatives have an unusually high ability to scavenge free radicals. (Synthesis of new lipophilic derivatives of fullerene C 60 / Sakhautdinov I.M., Islamova A.F., Mananov T.A., Gumerov A.M. // Bulletin of the Bashkir University. 2021. - T. 26. - No. 3. P. 650- 654). Experiments on animals show that fullerene compounds significantly increase their resistance to oxidative stress (Malikova RN, Sakhautdinov I.M., Ishbaeva SM, Yunusov MS Fullerene C60 DerivativesasEfficientSensitizersofOxidationundertheMildConditionsofAtmosphericAir // Russ. J. of Gen. Chem. 2017. - Vol. - 87. - No. 10. - P. 2497-2499).
2019. Т. 93. №11. С. 1674-1680).Due to the presence of π-conjugation in the C60 fullerene molecule, its electronic system is electron-deficient. For this reason, fullerene and its derivatives are capable of exhibiting the properties of a free radical scavenger, have a high radical capacity and can act as antioxidants in various systems, including biological ones, as well as compounds that can influence free radical processes and energy transfer processes. Along with antioxidant properties, fullerenes are able to transfer photoexcitation energy and thereby transfer oxygen from the triplet state to the singlet state (Antioxidant properties of amino acid derivatives of fullerene C 60 // Volkov V.A., Yamskova O.V., Shepel N.E., Romanova V.S., Kurilov D.V., Tregubov A.V., Vyshivannaya O.V., Voronkov M.V., Yamskov I.A., Misin V.M., Zubareva N.D., Kustov L.M. / / Journal of Physical Chemistry, 2019, Vol. 93, No. 11, pp. 1674-1680).

Resveratrol - 25 mg.
Alpha lipoic acid - 15 mg.
The drug was administered subcutaneously to white rats.
Полученный раствор оказал существенное воздействие на состояние белково-азотистого обмена и мочевинообразовательной функции печени, которую оценивали по динамике в крови общего белка, ферментов трансаминирования, а также метаболитов орнитинового цикла и системы глутаминовая кислота-глутамин (таблица 2).The resulting solution had a significant effect on the state of protein-nitrogen metabolism and urea-forming function of the liver, which was assessed by the dynamics in the blood of total protein, transamination enzymes, as well as metabolites of the ornithine cycle and the glutamic acid-glutamine system (Table 2).
After administration of the studied compound, the content of total protein, urea, ammonia and ornithine increased by 12.9%, 21.1%, 15.4% and 17.8%, respectively, compared to the control (Table 2). There were no significant differences in the glutamine content in the blood serum before and after administration of the studied compound.
The amino acid products of endogenous and exogenous protein digestion are broken down by hepatic transamination and oxidative deamination to form ammonia. Ammonia is then converted to urea by five enzymes in the urea cycle and excreted by the kidneys. The positive effect of the studied compound on protein-nitrogen metabolism in the body has been established.
После введения антиоксидантного препарата для животных на основе водного раствора фуллерена С60, ресвератрола и альфа-липоевой кислоты повышается и активность ферментов трансаминирования (фигура 2 и 3).After the administration of an antioxidant drug for animals based on an aqueous solution of fullerene C60 , resveratrol and alpha-lipoic acid, the activity of transamination enzymes also increases (Figures 2 and 3).
Transaminases are generally released at a constant rate, with normal levels in healthy animals representing a balance between normal hepatocyte turnover by programmed cell death (apoptosis) and enzyme clearance from the plasma.
Alanine aminotransferase (ALT) is an enzyme that catalyzes the transfer of amino groups to form the hepatic metabolite oxaloacetate. It consists of 496 amino acids, which are encoded by a gene located on the long arm of chromosome 8. ALT is found in abundance in the cytosol of hepatocytes. ALT activity in the liver is approximately 3000 times higher than that in serum.
In the event of hepatocellular injury or death, ALT release from damaged liver cells increases measured serum ALT activity, which is liver specific. ALT is also found in the kidneys and, in much smaller quantities, in heart and skeletal muscle cells.
The content of ornithine in the blood after administration of the studied compound increased by 17.8%, which is a favorable prognostic sign. Since the urea breakdown cycle occurs partly in the cytosol and partly in the mitochondria. For each revolution of the cycle, two molecules of nitrogen and one molecule of carbon dioxide are consumed. In return, one molecule of urea is created and one molecule of ornithine is regenerated to stimulate another round of the cycle. Beginning and ending with ornithine, the cycle reactions consume three equivalents of adenosine triphosphate and a total of four high-energy nucleotide phosphates; urea is the only new compound formed in the cycle.
Установлено, что после введения антиоксидантного препарата для животных водного раствора фуллерена С60, ресвератрола и альфа-липоевой кислоты активность аргиназы повышается на 26% по сравнению с контрольными животными (фигура 4).It was found that after the administration of an antioxidant drug for animals, an aqueous solution of C60 fullerene , resveratrol and alpha-lipoic acid, arginase activity increases by 26% compared to control animals (Figure 4).
The activity of lactate dehydrogenase (Figure 5), then after the introduction of an antioxidant drug for animals, an aqueous solution of fullerene C 60 , resveratrol and alpha-lipoic acid, did not change.
The concentration of total protein after the administration of an antioxidant drug for animals based on an aqueous solution of fullerene C60 , resveratrol and alpha-lipoic acid increased by 6.2% relative to control animals (Table 3). The level of albumin in the blood increased by 13.1%. The concentration of globulins after administration of the studied compound did not change relative to the control.
Thus, an antioxidant drug for animals based on an aqueous solution of C60 fullerene, resveratrol and alpha-lipoic acid has a pronounced antioxidant effect.
3.Example 3.

5.The drug was administered subcutaneously and intragastrically to white rats in doses of 500, 1000, 2500, 5000 mg/kg of the active substance. The animals were observed for 14 days after drug administration. The volume of solution administered to white rats was up to 3 ml. The research results are shown in tables 4 and 5.
Клиника токсического действия при подкожном введении антиоксидантного препарата для животных на основе водного раствора фуллерена С60, ресвератрола и альфа-липоевой кислоты у белых крыс, сопровождалась следующими симптомами (таблица 6).The clinical toxicity of the subcutaneous administration of an antioxidant drug for animals based on an aqueous solution of fullerene C60 , resveratrol and alpha-lipoic acid in white rats was accompanied by the following symptoms (Table 6).
Animals that received an antioxidant drug for animals based on an aqueous solution of fullerene C 60 , resveratrol and alpha-lipoic acid in the studied doses during the entire observation period (14 days) actively moved around the cages; in eating and drinking behavior they differed little from rats in the control group, adequate reaction to external stimuli (sound, light) was at the control level.
На вторые сутки после введения препарата восстанавливалось пищевое и питьевое поведение, реакции на внешние раздражители (таблица 7).When the drug was administered intragastrically at a dose of 2500 and 5000 mg/kg, white rats were depressed, inactive, and refused food during the first day. Reaction to external stimuli was reduced. On the second day after administration of the drug, eating and drinking behavior and reactions to external stimuli were restored (Table 7).
During 14 days of research, no deaths of experimental animals were noted. In this regard, it was not possible to establish the average lethal dose ( LD50 ). This allows us to consider an antioxidant drug for animals based on an aqueous solution of C60 fullerene, resveratrol and alpha-lipoic acid, non-toxic in the tested range and vary its doses within a wide range.

мг: водный раствор фуллерена С60 - 2 мл, 2 мг по действующему веществу, ресвератрол - 25 мг, альфа-липоевая кислота - 15 мг (таблица 10).Rabbits of the 1st experimental group were given an antioxidant preparation for animals based on an aqueous solution of fullerene C60 , resveratrol and alpha-lipoic acid, with the following ratio of components in May. mg: aqueous solution of fullerene C 60 - 2 ml, 2 mg of active substance, resveratrol - 25 mg, alpha-lipoic acid - 15 mg (Table 10).
Rabbits of the 2nd experimental group were administered an antioxidant preparation for animals based on an aqueous solution of fullerene C60 , resveratrol and alpha-lipoic acid. The animals were injected with an aqueous solution of fullerene C 60 - 2 ml, 2 mg of the active substance, resveratrol - 250 mg, alpha-lipoic acid - 150 mg.
It was established that application of solution 1 and 2 to the skin did not have a pathological effect on the skin of rabbits on both the 7th and 14th days of the experiments.
A drug based on an aqueous solution of fullerene C 60 for the prevention and treatment of free radical pathology in animals does not cause pronounced morphological changes in the various layers of the skin of experimental animals.
Guinea pigs were used to perform the conjunctival test. Guinea pigs of the 1st experimental group were treated with an antioxidant preparation for animals based on an aqueous solution of fullerene C60 , resveratrol and alpha-lipoic acid, with the following ratio of components in wt. mg: aqueous solution of fullerene C 60 - 2 ml, 2 mg of active substance, resveratrol - 25 mg, alpha-lipoic acid - 15 mg.
Guinea pigs of the 2nd experimental group were administered an antioxidant drug for animals based on an aqueous solution of fullerene C60 , resveratrol and alpha-lipoic acid. The animals were injected with an aqueous solution of fullerene C 60 - 2 ml, 2 mg of the active substance, resveratrol - 250 mg, alpha-lipoic acid - 150 mg.
The prepared solutions, 1 drop each, were injected with an eye pipette under the upper eyelid of the right eye, and distilled water was injected into the left eye, which served as a control. To evaluate the allergenic effect of each drug, 10 guinea pigs, 5 animals in groups, were used.
Immediate hypersensitivity was assessed 15 minutes after drug administration. Delayed-type hypersensitivity was assessed after 24-48 hours. The research results (Table 8) were assessed using the following scale:
1 - легкое покраснение слезного протока;1 - slight redness of the tear duct;
2 - покраснение слезного протока и склеры в направлении к роговице;2 - redness of the tear duct and sclera towards the cornea;
3 - redness of the entire conjunctiva and sclera. The reaction is accompanied by itching and when scratching with the paws, purulent ophthalmitis may develop.
During 6 hours of continuous observation of the animals, no changes in the clinical condition of the guinea pigs' body were recorded after applying the drug to the conjunctiva (Figure 8). According to the method, after a 24-hour and 48-hour observation period, no changes were recorded in the condition of the guinea pigs.
