Abstract
Description
Aliivibrio are bioluminescent gram-negative microorganisms, facultative anaerobes that colonize some marine animals and are also commonly found in seawater. Some strains of bacteria of the genus Aliivibrio are widely used for research in the field of bioluminescence, quorum sensing, as well as for the study of symbiotic microorganism-host mechanisms.
Aliivibrio is cultivated on many natural and synthetic media at a temperature of 20-25°C. After 24 hours, the culture forms small translucent colonies on solid media, or a wall ring, turbidity of the medium and the formation of a film on liquid media. The intensity and general presence of bioluminescence varies depending on the composition of the nutrient medium.
The optimal nutrient medium for the cultivation of Aliivibrio microorganisms in terms of price and availability was considered to be LB medium with the addition of 1.7% NaCl [2], however, after 1 day of cultivation at a temperature of 20°C, the number of bacterial colonies or the intensity of bioluminescence were often insufficient for further experimentation. A medium for growing bioluminescent bacteria is also known [1], however, even when grown on this medium, the luminescence of Aliivibrio does not manifest itself to the required extent.
It was found that when 2% sucrose was added to LB medium, after 18 hours of cultivation at a suitable temperature, Aliivibrio showed sufficient growth and bright bioluminescence.
The Aliivibrio logei culture was sown on Petri dishes with a medium prepared according to the source [1], source [2], and according to the proposed recipe. The dishes were placed in a thermostat with optimal cultivation conditions, and after 24 hours the intensity of bioluminescence was assessed.
1.The results are presented in Table 1.
As can be seen from the presented results, the proposed medium formulation provides a higher level of luminescence than known compositions.
