FIELD OF TECHNOLOGY TO WHICH THE INVENTION RELATES
The present invention is directed to improving the reaction medium for enzymatic synthesis reactions. The reaction medium contains a certain surfactant, which enhances the dissolution of the reactants and products of the chemical reaction and increases the activity of the enzyme compared to conventional surfactants. The use of a certain surfactant leads to an increase in yield and a more stable enzymatic reaction. For this purpose, the invention provides a reaction mixture containing an enzyme and a surfactant, and a method for carrying out an enzymatic reaction using said reaction mixture.
BACKGROUND OF THE INVENTION
New methods of catalytic synthesis in organic chemistry, meeting increasingly stringent environmental requirements, are in great demand in the pharmaceutical and chemical industries. In addition, new catalytic procedures are needed to obtain new classes of organic compounds that are becoming objects of molecular and biomedical research. Enzyme-catalyzed chemical transformations are now widely recognized as practical alternatives to traditional (non-biological) organic synthesis and as convenient solutions to some of the intractable problems of synthesis.
Enzymatic reactions are generally performed in an aqueous medium. However, many substrates and products of interest to the chemical industry are only partially soluble in water. Organic solvents are often added to the reaction mixture to improve solubility. On the other hand, these organic solvents adversely affect the stability and efficiency of many enzymes. In addition, some organic solvents are harmful to the environment.
Therefore, there is a need in the art for a reaction medium for enzymatic synthesis reactions that would allow the dissolution of hydrophobic substrates and products, but would not affect the stability and yield of the enzymatic reaction.
BRIEF DESCRIPTION OF THE PRESENT INVENTION
The present invention is based on the discovery that the efficiency of chemical synthesis reactions using enzymes as catalysts can be increased by introducing certain surfactants into the reaction mixture. Surfactants can be used in enzymatic synthesis reactions to increase the solubility of reactants and reaction products. However, these surfactants adversely affect the activity and stability of enzymes. The present inventors have found that the use of surfactants derived from vitamin E, such as TPGS-750-M, significantly improves the efficiency of the enzymatic reaction compared to conventional surfactants or co-solvents. The yield of the reaction also increases at lower temperatures and the reaction still proceeds even under unfavorable conditions such as low pH values.

In particular, it can be used for the synthesis of organic compounds using enzymatic reactions. Resistant enzymes for use in the present reaction mixture include, for example, oxidoreductases, transferases, hydrolases, lyases (synthases), isomerases, and ligases (synthetases). Oxidoreductases can catalyze the reaction of C=O and C=C bonds, reductive amination of C=O groups, and oxidation of C-H, C=C, C-N and C-O bonds. Transferases are capable of transferring functional groups such as amino, acyl, phosphoryl, methyl, glycosyl, nitro, and sulfur-containing groups from one compound to another. Hydrolases can catalyze the hydrolysis of esters, amides, lactones, lactams, epoxides, nitriles, and other groups, as well as the corresponding back reactions to form such functional groups. Lyases, also called synthases, can catalyze the addition of small molecules at double bonds such as C=C, C=N and C=O. Isomerases can catalyze the conversion of isomers (isomerization), including racemizations, epimerizations, and rearrangement reactions. Ligases, also called synthetases, can form complex compounds (similar to lyases) using ATP as an energy source. In specific embodiments, the enzyme is selected from the group consisting of ketoreductases, en-reductases, transaminases, dehydrogenases such as alcohol dehydrogenases and amino acid dehydrogenases, lipases, esterases, and phenylanine ammonium lyases. Specific examples of enzymes include ketoreductase, en-reductase, and alanine transaminase.
The enzyme is present in the reaction mixture at a concentration suitable for carrying out the reaction, for example, its content is from about 0.01% to about 100% relative to the content of the substrate. In particular, the enzyme content may be from about 0.1% to about 75%, from about 0.5% to about 50%, from about 1% to about 40%, from about 2% to about 30%, from about 4 % to about 25% or from about 5% to about 20% relative to the content of the substrate. "Content" in this respect means either the mass content of the compounds or the mole content of the compounds.
In certain embodiments, the aqueous reaction mixture further comprises a cofactor and/or a coenzyme. The presence of the cofactor/coenzyme depends on the enzymatic reaction to be carried out. Examples of cofactors include nicotinamide adenine dinucleotide (NAD), nicotinamide adenine dinucleotide phosphate (NADP), flavin adenine dinucleotide (FAD), and pyrodoxal monophosphate. These cofactors can be used to provide protons, electrons and/or amino groups in an enzymatic reaction. NAD, NADP and FAD may be present in the aqueous reaction mixture in ionic (eg NAD + , NADP + ) or protonated (eg NADH, NADPH) form.
The cofactor may be present in the aqueous reaction mixture in stoichiometric amounts. In particular, the molar content of the cofactor may be at least not less than the mole content of the substrate. "Content" in this respect means either the mass content of the compounds or the mole content of the compounds. In these embodiments, it is preferred that the cofactor be renewed during the course of the reaction, if necessary. For example, the reaction mixture may contain an additional enzyme, optionally with its corresponding substrate, which renews the cofactor. For example, if NAD, NADP, or FAD is used as the cofactor, the aqueous reaction mixture may further comprise a dehydrogenase such as alcohol dehydrogenase or glucose dehydrogenase and an appropriate substrate such as alcohol or glucose.
In other embodiments, the cofactor may be renewable by an enzyme that also catalyzes the reaction of interest. For example, for a transaminase reaction, the aqueous reaction mixture may contain an amine compound such as isopropylamine to regenerate a cofactor such as pyridoxal monophosphate.

The use of surfactants in biocatalysis to modify enzyme activities gives favorable results, such as an increase in the solubility of the reagents and an increase in the selectivity of the reaction. Although a number of surfactants, including ionic liquids, SDS and Triton X, have been widely used in bioconversion to date, the use of TPGS-750-M, developed in the Lipshutz laboratory, as an alternative surfactant has not been explored. During the investigation of the ketoreductase-mediated (KRED) reaction shown in Scheme 1, considerable effort was devoted to finding a suitable reaction medium, which was due to the low solubility of both starting ketone 1 and alcohol product 2 in this heterogeneous reaction.
DMSO.Initially, several co-solvent systems were used, including TPGS, PEG400 and DMSO, in aqueous solutions while screening for the above reaction conditions in parallel. As shown in FIG. 1, during the first phase of the reaction, which lasted 2.5 hours, the TPGS performance was better than that of PEG400 and DMSO, while the difference in conversion levels was 10%. As the reaction proceeded further, the reaction proceeded significantly faster in TPGS, and the observed difference in conversion levels was approximately 40% compared to the reaction in PEG400 and DMSO over a period of 6 hours. After 22 hours, the conversion in the reaction in TPGS was almost complete (100% ), while the reaction in PEG400 and DMSO stopped at 82% and 86% conversions, respectively. This fact is a consequence of the lower stability of ketoreductase in the presence of additions of organic solvents in the system of both PEG400 and DMSO.
With the screening results at our disposal, we were able to further optimize the response on a gram scale. We especially compared the performance of TPGS and DMSO systems. Thus, the reactions were carried out in 2% TPGS and 15% DMSO aqueous solution, respectively, at 40 ° C. using a 5 wt. % enzyme loading. The reaction reached 93% conversion after 18 h in the TPGS system, while a lower 80% conversion after 20 h was observed in the DMSO system. More importantly, the reaction proceeded and reached 98.8% conversion to TPGS after an additional 25 hours. And in the case of DMSO, an additional 2 wt.% enzyme was required to achieve 98.6% conversion (entry 1, table 1). This observation clearly demonstrates the higher level of activity and stable nature of the enzyme in TPGS compared to DMSO. Another interesting observation was that after unscheduled exposure of the enzyme for 24 h to pH 4.7 in the TPGS reaction system and after that pH was adjusted to 7, the enzyme was still active enough to further catalyze the reaction with the same activity level (record 2, table 1). Excellent stability can be obtained by molecular interactions between TPGS and protein and protection of the catalytically active site of the enzyme from the aqueous reaction medium.

Полученную суспензию отфильтровывали при 40oC, а полученный на фильтре осадок промывали и сушили с получением продукта 2 в виде грязно-белого твердого вещества (2,8 г, чистота 97%, выход 85%).Finally, the TPGS reaction is more suitable for scaling up and Compound 1 can be added in solid form without prior solution or milling to reduce particle size. A typical experimental procedure is as follows: to a degassed 2% by weight solution of TPGS-750-M in buffered water (34 ml, 10 vol.; standard solution prepared from 74 ml of a 2% by weight aqueous solution of TPGS-750-M, 1. 6 g Na 2 HPO 4 .12H 2 O and 0.5 g NaH 2 PO 4 .2H 2 O) were added glucose (3.2 g, 18 mmol, 2.0 eq.) to a mechanically stirred reactor equipped with a control system pH/ORP at RT The suspension was stirred at rt. for 20 minutes and to the resulting mixture were added NADP (59 mg, 68% purity), GDH (33 mg) and ketoreductase (0.16 g, 5 (wt.)% of substrate 1) in succession. Substrate 1 (3.3 g, 9 mmol, 1.0 eq.) was then added and the pH of the reaction mixture was adjusted to 6.8-7.2 with 1M aq. NaOH at RT The resulting mixture was heated to 40°C and stirred at 40°C for 43 hours until completion of the reaction, which was determined using HPLC. As the reaction progressed, product 2 precipitated from the reaction mixture and formed a suspension. The resulting suspension was filtered at 40 ° C. and the filter cake washed and dried to give product 2 as an off-white solid (2.8 g, 97% purity, 85% yield).
