Липидная оболочка таких систем обеспечивает инкапсуляцию и высокую клеточную проницаемость терапевтических агентов, а порфирины - их контролируемое выделение в клеточную среду за счет разрушения липосом в результате фотовозбуждения ковалентно-связанных порфириновых фрагментов под воздействием внешнего электромагнитного излучения. The lipid shell of such systems provides encapsulation and high cell permeability of therapeutic agents, while porphyrins ensure their controlled release into the cellular environment due to the destruction of liposomes as a result of photoexcitation of covalently bound porphyrin fragments under the influence of external electromagnetic radiation. This approach allows for the local therapeutic effect of drugs and chemotherapeutic drugs, minimizing possible side effects on the body as a whole.

Первым патентованным препаратом-фотосенсибилизатором явился фотофрин II, хорошо зарекомендовавший себя при ФДТ различных злокачественных новообразований и являющийся самым распространенным в мире фотосенсибилизатором в настоящее время. Полным аналогом фотофрина II в России является первый отечественный фотосенсибилизатор фотогем, который представляет собой смесь мономерных и олигомерных производных гематопорфирина. При поглощении фотогем способен переходить в возбужденное состояние и затем либо флуоресцировать в красной области спектра, либо вызывать фототоксичные реакции в опухолевой ткани. Указанные свойства позволяют использовать его как для диагностики опухоли, так и для ее удаления [Гейниц А.В., Цыганова Г.И. В настоящее время проводится направленный поиск таких фотосенсибилизаторов, обеспечивающих эффективную генерацию синглетного кислорода в дальней красной и ближней ИК-области 750-1500 нм (где находится максимальная проницаемость тканей), среди производных хлоринов, бактериохлоринов, пурпуринов, бензопорфиринов, тексафиринов, этиопурпуринов, нафтало- и фталоцианинов. При этом особый интерес представляют фотосенсибилизаторы, обладающие способностью не только быстро накапливаться в опухолях, но и с высокой скоростью распадаться. За последние 10 лет была разработана технология извлечения из растительного сырья комплекса биологически активных хлоринов, которые содержат в качестве основного компонента хлорин-е6. В результате были созданы фотосенсибилизаторы второго поколения - фотохлорин и фотодитазин [RU 2276976 С2, 27.05.2006]. Последние способны разрушать биологические субстраты после возбуждения светом с длиной волны в диапазоне 654-670 нм, чему соответствует эффективная глубина проникновения света до 7 мм вглубь тканей. Препараты имеют высокую степень фототоксичности, связанную с высоким квантовым выходом синглетного кислорода. Сохраняющаяся при этом способность препаратов флуоресцировать оставляет возможность для люминесцентной диагностики очагов неопластического изменения тканей. In this case, of particular interest are photosensitizers that have the ability not only to rapidly accumulate in tumors, but also to decompose at a high rate. Over the past 10 years, a technology has been developed for extracting a complex of biologically active chlorins from plant raw materials, which contain chlorin-e6 as the main component. As a result, second-generation photosensitizers were created - photochlorin and photoditazine [RU 2276976 C2, 27.05.2006]. The latter are able to destroy biological substrates after excitation by light with a wavelength in the range of 654-670 nm, which corresponds to an effective depth of light penetration up to 7 mm deep into tissues. The preparations have a high degree of phototoxicity associated with a high quantum yield of singlet oxygen. The ability of preparations to fluoresce, which remains, leaves the possibility for luminescent diagnostics of foci of neoplastic tissue changes.
