Abstract
Description
The invention relates to the chemical and pharmaceutical industry and can be used in quality control centers for medicines and control and analytical laboratories when analyzing the content of syringin in the bark of common lilac.
The current quality control system requires continuous improvement of approaches to the standardization of biologically active compounds (BAS) using modern methods of analysis and up-to-date data on physicochemical, spectral and pharmacological properties, which make it possible to differentiate specific BAS.
(2, 5). Common lilac ( Syringa vulgaris L., family Oleaceae ) is a deciduous shrub, cultivated as an ornamental plant, but known for its medicinal properties (1-4). During a phytochemical study by scientists of the Samara State Medical University and VILAR, it was revealed that the dominant biologically active compound of the lilac bark is cinnamyl glycoside syringin (2,3,5). In official medicine, common lilac bark is used as a raw material for obtaining the state standard sample of eleutheroside B (syringin) (VFS 42-2088-92 "Siringin-standard sample"), used in assessing the quality of rhizomes and roots of Eleutherococcus senticosus Maxim, a family . Araliaceae - Araliaceae ), drugs and dietary supplements based on raw materials of Eleutherococcus senticosus (2, 5).
Для определения содержания сирингина в коре сирени используются спектрофотометрические, хроматоспектрофотометрические методы, метод высокоэффективной жидкостной хроматографии (ВЭЖХ) (2,5).To determine the content of syringin in lilac bark, spectrophotometric, chromatospectrophotometric methods, and high performance liquid chromatography (HPLC) are used (2.5).
(6): As a prototype, we took the pharmacopoeial method for determining syringin in the rhizomes and roots of Eleutherococcus senticosus (6). The method involves fractional extraction of syringin using 96% ethanol as an extractant under reflux in a water bath. HPLC analysis of syringin content is carried out under the following conditions (6):
- колонка из нержавеющей стали 250×4,6 мм, эндкепированный октадецилсиликагель для хроматографии, диаметр 5 мкм, - stainless steel column 250×4.6 mm, endcapped octadecyl silica gel for chromatography, diameter 5 µm,
- элюирование в градиентном режиме, подвижная фаза – раствор фосфорной кислоты концентрированной в воде и ацетонитрил для хроматографии в разных соотношениях.- elution in a gradient mode, the mobile phase is a solution of concentrated phosphoric acid in water and acetonitrile for chromatography in different ratios.
- detection at a wavelength of 266 nm.
The duration of the analysis can be up to 45 minutes.
(2,3).In our opinion, the extraction parameters used in the prototype do not allow for a high yield of syringin, since syringin is slightly and slowly soluble in 95% ethanol (2,3).
(7).In addition, it is known that in the gradient mode, the adjustment of conditions is more critical, it can lead to incorrect identification of peaks, their overlap or shifts, in which substances of interest can exit after the specified time of registration of the chromatogram (7).
Thus, the aim of the invention is to develop a new method for determining the quantitative content of syringin in the bark of the common lilac with an increased yield of syringin and a shorter duration of analysis while maintaining the correctness, specificity and precision of the method.
The technical result is an increase in the yield of syringin and a decrease in the duration of the method for the quantitative determination of syringin in the bark of common lilac.
The technical result is achieved by the fact that the method is carried out by extracting syringin from the bark of common lilac with 70% ethyl alcohol, HPLC analysis of the obtained extracts in isocratic elution mode using a mixture of acetonitrile and 1% acetic acid solution in water as a mobile phase in a ratio of 15:85 and detection at a wavelength of 266 nm.

Извлечение фильтруют через бумажный фильтр (раствор А). 5 мл полученного раствора помещают в мерную колбу вместимостью 25 мл, доводят водой очищенной до метки (раствор В).The method is carried out as follows. An analytical sample of raw materials is crushed to the size of particles passing through a sieve with holes with a diameter of 1 mm. About 1 g of crushed raw material (accurately weighed) is placed in a flask with a thin section with a capacity of 100 ml, 30 ml of 70% ethyl alcohol is added. Stopper the flask and weigh on a balance to the nearest ±0.01 g. Attach the flask to a reflux condenser and heat in a boiling water bath (moderate reflux) for 60 minutes. Then the flask is cooled for 30 min, closed with the same stopper, weighed again, and the missing extractant is replenished to the original weight. The extract is filtered through a paper filter (solution A). 5 ml of the resulting solution is placed in a volumetric flask with a capacity of 25 ml, adjusted with purified water to the mark (solution B).
1 µl of solution B is injected into a Milichrom-6 liquid chromatograph with a UV detector. Chromatographed under conditions of reversed-phase chromatography in isocratic mode: steel column "KAH-6-80-4" (2 mm x 80 mm; Separon-C18, diameter 5 μm), mobile phase - acetonitrile: 1% solution of acetic acid in water in a ratio of 15:85, elution rate - 100 µl/min, eluent volume - 1000 µl.
Conduct UV detection at a wavelength of 266 nm, the sensitivity range of 0.5. Conduct at least 3 parallel determinations.
In parallel, 1 µl of the syringin standard solution is injected into the chromatograph and chromatographed as described above. The height of the syringin peak is determined and the average peak height is calculated from the results of three parallel determinations.

Note: Preparation of syringin standard sample solution. About 0.025 g (accurately weighed) of the state standard sample of syringin (content of the main substance ≥95%) is placed in a volumetric flask with a capacity of 50 ml, dissolved in a small amount of 95% ethyl alcohol when heated in a water bath, brought to the mark with 95% ethyl alcohol, mixed .
The validation evaluation of the developed methodology was carried out according to the following indicators: specificity, linearity, correctness (openness), precision. The specificity of the method was determined by the correspondence between the retention times of the standard sample of syringin and the corresponding peak on the HPLC chromatogram of the test solution, as well as by the resolution between the closest peaks and the asymmetry factor of the syringin peaks.
In FIG. 1 shows the HPLC chromatogram of a standard solution of syringin, FIG. 2 – HPLC-chromatogram of water-alcohol extract from common lilac bark (symbols: 1 – water-alcohol extract from common lilac bark; 2 – standard sample of syringin). Under the conditions of HPLC analysis, the retention time of the syringin peak on the chromatograms of the standard sample of syringin and water-alcohol extract from the bark of common lilac was 4.046±0.070 min and 4.092±0.082 min, respectively.
Linearity was determined at five concentration levels of syringin solutions (with concentrations ranging from 0.3424 to 0.5136 mg/ml). The correlation coefficient was 0.99971. This range can be considered as the analytical area of the technique.
98,8 %. The correctness of the technique was determined by the method of additions by adding a solution of syringin with a known concentration (80%, 100% and 120%) to the test solution. At the same time, the average open rate was 98.8%.
1,20 %.The study of the internal precision of the method indicates the convergence of the obtained concentrations of the analyte: the error in determining the average result of the content of syringin in the bark of common lilac with a confidence probability of 95% was ±3.20%. When evaluating the intralaboratory precision, satisfactory results were also shown, since the relative error in the determination of syringin on the first and second days of analysis is in the range from 0.82% to 1.20%.
В результате эксперимента было определено, что оптимальными параметрами экстракции являются: однократное извлечение 70 % этиловым спиртом на кипящей водяной бане в течение 60 минут в соотношении «сырье-экстрагент» - 1:30.We also studied the effect of the extractant on the extraction process. Table 1 shows the effect of various extraction parameters (extractant, extraction duration, raw material-extractant ratio) on the completeness of syringin extraction from common lilac bark. As a result of the experiment, it was determined that the optimal extraction parameters are: a single extraction with 70% ethyl alcohol in a boiling water bath for 60 minutes in the ratio of "raw material-extractant" - 1:30.
Taking into account that an increase in the number of operations at the stage of sample preparation leads to an increase in the error, the choice was made in favor of a single-stage extraction process with confirmation of the required accuracy of quantitative determination.
The proposed method is illustrated by the following example.
Example.
Извлечение фильтруют через бумажный фильтр (раствор А). 5 мл полученного раствора помещают в мерную колбу вместимостью 25 мл, доводят водой очищенной до метки (раствор В).An analytical sample of raw materials is crushed to the size of particles passing through a sieve with holes with a diameter of 1 mm.
The content of syringin in the bark of common lilac in terms of absolutely dry raw materials in percent ( X, % ) is calculated by the formula
где H = 472,7941 – среднее значение высоты пика сирингина, вычисленное из хроматограмм раствора испытуемого образца;where H = 472.7941 is the average height of the syringin peak calculated from the chromatograms of the test sample solution;
Hо = 327,3610 – среднее значение высоты пика сирингина, вычисленное из хроматограмм раствора стандартного образца сирингина;H o = 327.3610 is the average value of the syringin peak height calculated from the chromatograms of the syringin standard sample solution;
m = 1,0510 – масса сырья, г;m = 1.0510 is the mass of raw materials, g;
mо = 0,0249 – масса стандартного образца, г;m o = 0.0249 is the mass of the standard sample, g;
W = 11 – потеря в массе при высушивании сырья, в процентах;W = 11 - weight loss during drying of raw materials, in percent;
5,01 % ± 0,18 %.The test was carried out in triplicate. All data were statistically processed. The content of syringin was 5.01% ± 0.18%.
Thus, the proposed method for the quantitative determination of syringin in the bark of common lilac has the following advantages:
1. Elution in isocratic mode avoids problems (incorrect peak identification, peak overlap, or shifts in which substances of interest can exit after the specified chromatogram acquisition time) that occur when performing chromatographic analysis in gradient mode.
2. The duration of the chromatographic analysis is 10 minutes compared to 45 minutes in the prototype.
The use of 70% ethanol in the developed method makes it possible to increase the extraction efficiency by 2 times (when comparing the results of the analysis of syringin when using 70% ethanol and 95% ethanol as extractants, as in the prototype) (table 1).
4. The error in determining the average result of the syringin content with a confidence probability of 95% is + 3.20%, which indicates a high reproducibility of the technique.
