Как известно, для живых клеток характерны локальные колебания мембраны, называемые также фликкер или динамические флуктуации мембраны. Несмотря на длительную историю исследования таких колебаний, и многочисленные работы, посвященные их спектральному анализу и моделированию, молекулярные и клеточные механизмы данного явления до сих пор обсуждаются. Важно, что не подвергается сомнению тот факт, что амплитуда фликкера определяется мембраной и зависит от состояния цитоскелета клетки: изменения упругих свойств комплекса мембраны и цитоскелета или формирования локальных изгибов (выступов) мембраны за счет перестройки спектриновой сети (например, отщепления от узлового комплекса одной из нитей спектрина и образования непланарной конфигурации сети). Также при этом возможно анализировать и характер шума, выделяя определенные частоты колебаний и оценивая с их помощью динамические процессы, протекающие в клетках. Данный процесс весьма сложен и требует значительных человеческих и машинных трудозатрат. Понимание происхождения клетки и определение характеристик колебания ее мембраны могут дать информацию о функциональных состояниях эритроцитов при нормальных и патологических состояниях. Не менее важной задачей является изменение плотности внутри клетки, связанные с синтезом и распадом белка, что также приводит к периодическим изменениям фазы, прошедшего через объект волнового фронта. Разделить влияние колебания мембраны и флуктуаций плотности внутри клетки из оптических измерений практически невозможно, поэтому определяют изменение оптической толщины клетки, характеризующей обе эти величины. Для исследования динамики живых клеток используются различные методы, такие как: фазово-контрастная микроскопия, отраженная интерференционная контрастная микроскопия, рассеяние света, техника, основанная на точечной темнопольной микроскопии. Understanding the origin of the cell and determining the characteristics of the oscillation of its membrane can provide information about the functional states of erythrocytes under normal and pathological conditions. An equally important task is to change the density inside the cell, associated with the synthesis and decay of protein, which also leads to periodic changes in the phase that has passed through the wavefront object. It is practically impossible to separate the influence of membrane oscillations and density fluctuations inside the cell from optical measurements; therefore, the change in the optical thickness of the cell, which characterizes both of these quantities, is determined. Various methods are used to study the dynamics of living cells, such as phase contrast microscopy, reflected interference contrast microscopy, light scattering, and a technique based on point dark-field microscopy. However, these methods are not inherently quantitative from the point of view of absolute measurements and do not allow one to study the distribution of membrane vibrations and fluctuations of the refractive index inside the cell.

Когерентная фазовая микроскопия внутриклеточных процессов // Успехи физических наук.- 2001. - Т. 171. - №6.). В качестве источника освещения использовался He-Ne лазер. Интерференционное изображение регистрируется с помощью координатно-чувствительного фотоприемника - диссектора, который представляет собой электронно-оптический преобразователь с внешним фотоэффектом (без накопления заряда) с фотоэлектронным умножителем и магнитным переносом потока электронов в плоскость диафрагмы с малым отверстием.
