Экзосомы - мембранные нано-везикулы (80-130 нм), которые секретируются практически всеми клетками многоклеточных организмов во внеклеточное пространство. Супернатант удалили, к осадку добавили 1,4 мл содержимого второй пробирки (раствор ПЭГ и декстрана в фосфатно-солевом буфере). Осадок ре-суспендировали, смесь центрифугировали при 1000 G, 10 минут, при +4°С. После удаления супернатанта, осадок содержащий экзосомы растворяли в лизис-буфере (реагент «Лира», Биолабмикс / Новосибирск). Далее проводилось выделение микроРНК с помощью колонок и реагентики в составе набора для выделения (LRU-100-50, Биолабмикс / Новосибирск) по протоколу производителя. Параллельно экзосомы были выделены из аналогичных образцов плазмы (1,5 мл) с использованием коммерческих наборов (ближайших аналогов), наиболее близких по принципу работы к заявленному способу: ExoPrep (Hansa Bio Med) и Tota lexosomes isolation kit (Invitrogen). Выделение экзосом проводилось в соответствии с протоколами производителей, микроРНК была выделена аналогичным образом с помощью реагента «Лира» и набора для выделения (LRU-100-50, Биолабмикс / Новосибирск). Во всех трех случаях элюцию РНК проводили 50 мкл буфера (ЕБ), для последующего анализа микроРНК использовали по 2 мкл раствора. С помощью ОТ-ПЦР был проведен анализ относительной концентрации трех молекул микроРНК (miR-29b-1, miR-451a и miR-375) с помощью ранее описанной технологии «two-tailed reverse transcription) [19, 20]. Isolation of exosomes was carried out in accordance with the manufacturer's protocols, microRNA was isolated in the same way using the Lira reagent and an isolation kit (LRU-100-50, Biolabmix / Novosibirsk). In all three cases, RNA was eluted with 50 μL of buffer (EB); for subsequent analysis of microRNA, 2 μL of solution was used. Using RT-PCR, the relative concentration of three microRNA molecules (miR-29b-1, miR-451a, and miR-375) was analyzed using the previously described “two-tailed reverse transcription” technology [19, 20]. All reactions were carried out in volumes of 20 μl, standard kits from Biolabmix were used: OT-M-MuLV-RH for reverse transcription and HS-qPCRSYBR Blue (2 ×) for PCR.
