Abstract
Description
The invention relates to medicine and biology, namely to the study of the rheological properties of blood and can be used to assess the state of aggregation of red blood cells.
A known photometric method for assessing red blood cell aggregation (Schmid-Schonbein et al., Microvasc. Res. 1973. Vol. 6. P. 366-376), including the isolation of red blood cells from blood, mixing with autoplasma or with high molecular weight dextran, placing a rheoscope in a cuvette and registration of the aggregatogram record.
Using the photometric method, you can determine the degree of aggregation of red blood cells in the form of “coin columns”. However, the intravital morphology of the aggregate state of erythrocytes with a large increase in the light microscope remains practically unstudied.
Known methods for studying the morphology of red blood cell aggregates in autologous plasma and blood serum with a large increase in light microscope (Sheremetyev Yu.A., Uspensky AN Method for microscopic evaluation of the state of aggregation of red blood cells in autoplasma. Patent RU 2605843, 2016; Sheremetyev Yu.A., Uspensky AN A method for assessing the state of aggregation of red blood cells in autologous blood serum. Patent RU 2590985, 2016). Due to the fact that high molecular weight polymers (dextran, polyvinylpyrrolidone) are widely used as aggregating agents both for studying the mechanisms of interaction of red blood cells and for assessing their aggregation ability in pathologies of various origins, it seems important to develop a method for morphological assessment of red blood cell aggregation induced by high molecular weight polymers , with a large increase in the light microscope.
The prototype is a method of microscopic assessment of the state of aggregation of red blood cells in an autoplasma (Sheremetyev Yu.A., Uspensky A.N. Method of microscopic assessment of the state of aggregation of red blood cells in an autoplasma. Patent RU 2605843, 2016), including blood sampling, isolation of red blood cells, mixing blood plasma with erythrocytes in a ratio of 2: 1, placing the mixture on a glass slide, mixing, lowering the lens mixture × 100 into a drop, microphotography.
(Wong P. A basis of echinocytosis and stomatocytosis in disc-sphere transformation of the erythrocyte. J. Theor. Biol. 1999. V. 196. P. 343-361). (Wong P. A basis of echinocytosis and stomatocytosis in disc-sphere transformation of the erythrocyte. J. Theor. Biol. 1999. V.196. P. 343-361). Normal red blood cells are known to transform into echinocytes when two glass surfaces come in contact (Wong P. A basis of echinocytosis and stomatocytosis in disc-sphere transformation of the erythrocyte. J. Theor. Biol. 1999. V. 196. P. 343-361) . This is called the “glass effect." The glass effect, in particular, occurs when a 100 x lens is lowered into a suspension of red blood cells placed on a slide (Sheremetyev Yu.A., Popovicheva A.N., Uspensky A.N., Aleksandrov A.A., Uspenskaya O. A. Method for intravital assessment of the action of reagents on the formation of stomatocytes. Patent RU 2629609, 2017). In the prototype method, the glass effect is eliminated due to the fact that the plasma contains albumin protein that prevents the transformation of cells into echinocytes (Wong P. A basis of echinocytosis and stomatocytosis in disc-sphere transformation of the erythrocyte. J. Theor. Biol. 1999. V.196. P. 343-361). Polymer solutions do not contain plasma proteins; therefore, upon contact of two glass surfaces in their presence, red blood cells are transformed into echinocytes.
The objective of the proposed invention is the creation of a simple, quickly implemented method of microscopic assessment of the state of aggregation of red blood cells in the presence of high molecular weight polymers.
The problem is solved due to the fact that a solution of high molecular weight polymer in the presence of 0.1% albumin is mixed with washed red blood cells in a 2: 1 ratio, the mixture is placed on a glass slide, mixed, the lens (× 100) of the light microscope is lowered into it and after reaching the focus (× 1000) is observed through the eyepiece of the microscope. For microscopic assessment of the state of aggregation of red blood cells, microphotography is performed.
The method is as follows. Blood is collected in vacuum tubes containing 3.8% sodium citrate (9: 1 ratio). The tubes are centrifuged at 3000 rpm for 20 minutes Plasma and white blood cells are removed, after which the red blood cells are washed three times with physiological saline. As high molecular weight polymers, a 3% solution of dextran 70 (m.v. 70 kDa, Sigma, USA) or a 0.5% solution of polyvinylpyrrolidone (m.v. 360 kDa, Sigma, USA) are used. Dextran or polyvinylpyrrolidone is prepared in buffered saline (10 mM Tris-HCl, 150 mM NaCl, pH 7.4) containing 0.1% albumin. A solution of dextran or polyvinylpyrrolidone containing 0.1% albumin is mixed with red blood cells in a 2: 1 ratio, a drop of the mixture is placed on a glass slide, mixed, the lens (× 100) of the light microscope is lowered into it and after reaching the focus (× 1000), microphotography is performed.
Figure 1 shows the formation of “coin columns” from the erythrocytes of healthy donors after adding 3% dextran solution containing 0.1% albumin to them. There is a lack of echinocytes.
Figure 2 shows the formation of “coin columns” from red blood cells from healthy donors after adding 0.5% solution of polyvinylpyrrolidone containing 0.1% albumin to them. There is a lack of echinocytes.
Thus, the proposed method allows morphological assessment of red blood cell aggregation with a large increase in the light microscope, not only in autologous plasma and blood serum, but also in a solution of high molecular weight polymer.
