Abstract
Description
The invention relates to veterinary medicine, and in particular to methods of treating diseases of the respiratory tract in horses. The invention can also be used for the treatment of respiratory diseases in other types of farm animals - cattle, sheep, as well as in non-productive animals - cats and dogs.
Among various diseases of non-contagious etiology, the share of respiratory system diseases in horses accounts for up to 70%. This prevalence, along with the low efficacy of therapeutic measures, determines the interest of veterinary specialists in finding new affordable pharmacocorrection tools for bronchopulmonary pathology in horses.
Among the therapeutic measures, the main place is given to the full feeding of animals and the creation of an optimal microclimate. Means etiotropic and pathogenetic therapy include steroid anti-inflammatory drugs, broad-spectrum antibiotics, antiprotozoal and fungicidal agents, bronchodilators and substances with a desensitizing effect. Such complex treatment is applied everywhere and contributes, as a rule, to the rapid onset of short-term remission.
If we consider the disease from the standpoint of violation of the barrier properties of the respiratory system, the therapeutic measures are not aimed at restoring natural protection, but rather the opposite.
At the same time, it is known that organ and tissue systems have their own arsenal of barrier properties. So, all over the respiratory organs are lined with mucous membrane, the goblet cells of which secrete bactericidal mucus, and the equipment of the receptor apparatus allows to remove large pathogenic particles outward with reflex movements. The presence of natural non-specific factors of immunity allows you to immediately respond to the introduction of a foreign agent through a complex interrelated chain of immune responses.
The basis of the nonspecific components of the immune response is the inflammatory reaction, the inductors of which are specific (bacterial, viral, fungal, mycoplasma, etc.) or the products secreted by them, as well as components of the damaged mucosa. An important role of this reaction is to attract to the place of introduction of foreign agents of cells of the immune system (eosinophils, macrophages, lymphocytes) and their activation. They secrete inflammatory mediators, incl. cytokines (interleukin-8), and contribute to the destruction or absorption of a foreign protein. During the non-specific reaction, a number of humoral defense systems are activated, as already noted.
Thus, it should be noted that the most important group of cells of the immune system are antigen-presenting cells and a population of phagocytes, the development and activation of which are influenced by some cytokines. In the lung tissue, these are alveolar macrophages that serve as intermediaries between the airspace of the alveoli and the bloodstream. Alveolar macrophages are capable of secreting lysozyme, prostaglandins, complement components, leukotrienes, regulatory peptides, and especially cytokines. By producing fibronectin and related proteins, macrophages are involved in the formation of the intercellular matrix, which is an essential component of the microenvironment of immune system cells.
In chronic obstructive diseases of the respiratory system in horses, especially with repeated inhalation contact with a harmful substance, a picture of secondary immunodeficiency arises due to increased suppressor mechanisms and polyclonal activation, which, as a rule, is not amenable to correction by traditional immunomodulators or inductors of cytokine synthesis due to depletion of compensatory immune systems system. An additional reason for the death of immunocompetent cells, in addition to the action of organo-aggressive microorganisms, fungi, allergens, is unreasonably frequent use of corticosteroids (a classic example of apoptosis of resting lymphocytes from steroid hormones, well studied in medicine).
Thus, the use of those immunotropic drugs that would eliminate the deficiency of a key component of an adequate immune response and at the same time contributed to the normalization of antigen recognition, proliferation and differentiation of immune cells, secretory activity of macrophages, in particular alveolar, seems to be quite reasonable.
One of these immunotropic drugs is Roncoleukin, obtained by modern methods of biotechnology and genetic engineering.
However, this drug gives a pronounced therapeutic effect only in the complex treatment of obstructive horse disease (Kryachko OV, Romanova OV. Use of the drug Roncoleukin in chronic diseases of the lungs in horses., Recommendations. St. Petersburg, 2004. 27 p.) .
The closest in technical essence is the drug Pantoin (P RU No. 2406511 dated 12/20/2012 and P RU No. 2476237 dated March 10, 2013), which is used to increase the effectiveness of vaccination and adaptive capacity in horses.
However, this drug has a weak immunomodulatory effect on alveolar macrophages in the organs of the respiratory system of horses and does not always lead to recovery.
The objective of the invention is to expand the Arsenal of tools and methods for the treatment of diseases of the respiratory system of horses, contributing to improving the effectiveness of treatment, reducing recovery time and reducing the cost of treatment of animals.
%:The problem is solved in that the preparation for treating diseases of the respiratory tract in horses, including an extract from reindeer antlers, a stabilizer, a filler according to the invention, additionally contains arabinogalactan (extract from Siberian larch), and physiological saline is used as a filler, in the following ratio components, mass. %:
The task is also solved by the fact that a method of treating diseases of the respiratory tract in horses, including the introduction of the drug according to the invention, injects a preparation containing an extract from reindeer antlers, arabinogalactan, nipagin, saline, in the following ratio, mass. %: extract from antlers of reindeer 25-30, nipagin 0.047-0.07, arabinogalactan (extract from Siberian larch) 5-10, saline up to 100, at a dose of 5 ml per head, 3-4 times, with an interval of 5- 7 days.
1. Preparation of the drug and determination of the effective therapeutic dose.
The drug was prepared according to the method of V.G. Shelepova and N.S. Osintsev (P RU No. 2008906 from 03.13.1994) by gradual extraction of reindeer antlers. The extract obtained was filtered, separated from the alcohol by evaporation in a water bath. After that, 5-10 arabinogalactan powder was added to the obtained extract and set for 30 minutes on an electronic mixer.
The resulting mixture was diluted with sterile saline to the original volume of the alcohol extract, the stabilizer nipagin (paraoxybenzoic acid methyl ester) was added. The vials were flawed, sealed with pre-flooding corks and metal caps under running-in, and sterilized with ultrasound.
The obtained preparation for biochemical and biological properties met the requirements as a tonic substance (biologically active additive). Immunostimulatory effect of which is enhanced by a combination of extracts of reindeer antlers and the use of arabinogalactan, a polysaccharide obtained from Siberian larch, which has a powerful immunomodulatory, antiseptic, probiotic effect on the animal's body.
При подборе компонентов препарата - экстракт пантов составлял 25-30 г на 100 мл готового препарата (таблица 1).When selecting the components of the drug - antlers extract was 25-30 g per 100 ml of the finished product (table 1).
The amount of arabinogalactan was selected empirically. Various doses of arabinogalactan (0.5-1.5 g; 2-3 g; 3-5 g, and 5-10 g) were added to the residue after evaporation in a water bath of the alcoholic extract, then the bottle with these contents was set for 30 minutes magnetic stirrer. After adding saline to 100 ml, the presence of a precipitate was observed. With a dose of arabinogalactan 0.5-1.5 g, a precipitate in the form of flakes remained at the bottom of the vial, as well at a dose of 2-3 g arabinogalactan, at a dose of 3-5 g, the precipitate was in the form of dust. In a bottle with 5-10 g of arabinogalactan sediment was not observed. The presence of sediment in vials with smaller doses of arabinogalactan means that the added doses are not enough to bind all the components of the extract of antlers, which because of this precipitate when adding a saline solution, as the antler extract is soluble only in alcohols.
The invention is illustrated by the following examples.
Example 1. Determination of the optimal dose of the drug and the frequency of administration.
To determine the dose of the proposed drug, 9 experimental and 1 control groups of horses were formed on the principle of analogues with 3 animals each (stallions and measures at the age of 8-10 years), all animals showed signs of chronic obstructive pulmonary disease (COPD): persistent chronic, dry , deep cough, severe shortness of breath.
Horses were kept in similar conditions (maintenance, exercise, feeding). Animals were injected with the proposed drug according to various regimens. Table 2 presents the different doses of the drug and the frequency of their introduction.
From the results shown in Table 1, it can be seen that the remission period increased with the administration of the drug in doses of 5 ml (once in 5 days 3 injections) and 10 ml (three times with an interval of 5 days and three times with an interval of 10 days). Since the achieved effect from the administration of these doses was similar, the optimal dose was selected for efficacy, frequency of administration, consumption of the drug, i.e. dose of 5 ml three times with an interval of 5 days. The total consumption of the drug was 15 ml. Side effects in all cases of the drug were absent.
2. Determination of the effectiveness of the drug in the treatment of respiratory diseases in horses.
To study the therapeutic efficacy of the drug, groups of horses were formed (n = 4), with symptoms of COPD, one control and three experimental (table 3). The test preparation was administered to the horses of the experimental groups three times with an interval of five days at a dose of 5 ml, the second group was given Ligfol and the third group Roncoleukin doses in doses recommended by the manufacturer. Animals of all groups were observed: they paid attention to the type of breathing (chest, abdominal), fatigue with mild athletic load, the presence of discharge from the nasal passages.
From the data given in Table 3, it can be seen that a stable therapeutic effect from the tested drug comes after the third injection. After the first injection, no obvious changes were observed. After the second injection, a decrease in the frequency of respiratory movements was noted. But the type of breathing remained abdominal, reducing fatigue at work — the horses could already bear a light load — a lynx 10 minutes. step 10 min., lynx 10 min., with auscultation, the whistling noises in the lungs disappeared, nasal discharge became mucous and practically stopped. After the third injection of the drug - abdominal breathing disappeared, no discharge of the nasal passages was observed, the animals normally maintained the usual load - lynx 15 minutes; step 5 min .; lynx 15 min; step 5 min .; 10 min. gallop.
3. Examples of treatment of horses with diseases of the respiratory system with the proposed drug in combination with other drugs.
Treatment of diseases of the respiratory system of horses is most often carried out in a complex, i.e. in combination with the most appropriate symptoms and methods for the symptoms. Table 4 shows the results of treatment with the proposed drug with the addition of a syrup containing potassium iodide, which contributes to a better separation of bronchial mucus.
Also, our proposed drug was used in combination with inhalations - eucalyptus, hay rot and others.
From the data given in Table 4, it can be seen that the greatest effect (remission up to 3.5 months) is achieved with a combination of injection of the drug and simultaneous evaporation of the syrup containing potassium iodide. With treatment with injection of the drug and inhalation in sports horses, the period of remission is the same with the use of only the drug. But for horses that do not carry a serious athletic load - the additional use of inhalation lengthens the remission period to 2.5 months.
In the treatment of pneumonia in young horses (up to 6 months of age), a standard regimen with antibiotics was used. In one group, the vitamin complex was replaced by the proposed drug. Recovery of animals of this group occurred faster (within two weeks). Animals of the first group recovered within four weeks. Examples of the practical application of the proposed drug.
1 example. Horses in the summer sports group, very hot climate, very dry air and a lot of dust - very bad weather for horses with chronic obstructive pulmonary disease. They poured syrup with potassium iodide, on the third day they began a course of the drug - 5 ml three times with an interval of 5 days. The remission period increased from 1.5 months to 3.5. Horses successfully performed the whole competition season.
2 example. Horses with COPD (amateur private traders) had a rolling load (without sports). The potassium iodide syrup was introduced (evaporated), on the third day the use of the proposed drug was started - 5 ml three times with an interval of 5 days. The remission period increased from 1.5 months to 3.5. Horses successfully worked the whole season.
3 example. Stallion - age 6 years. She works in hippotherapy, stood on a stable, where there were horses with symptoms of COPD. A few days later the animal had a cough, very strong, with seizures. After the first injection - the next day, there was no cough; a course of drug treatment was given - 5 ml three times with an interval of 5 days. Cough was not diagnosed, the condition remained stable.
4 example. The horses of 12 heads were diagnosed with pneumonia, the symptoms were wheezing, wheezing with crepitus on auscultation, temperature 40.5-41.3, purulent outflows from the nose, general depression. They were divided into two groups — the first group — 5 heads — were prescribed a course of antibiotics, autohemotherapy, and the 5 ml drug we proposed, every 5 days. The second group - 7 goals - was prescribed standard treatment - antibiotics, vitamin therapy, aloe vera, autohemotherapy. By the end of the third week, the horses of the first group were already trotting, with auscultation the noises in the lungs almost disappeared. In the horses of the second group, improvement in the condition came two weeks later, by the end of the third week after the lynx - shortness of breath, the horses sweat.
Thus, our proposed drug can be recommended for maintenance therapy of horses with symptoms of COPD. The use of this drug will significantly increase the possibility of operating sport horses. It will help them to hold the competitive season in a stable working form. It is also recommended for horses of a hobby class (horses of private owners) to maintain a healthy state.
