Abstract
Description
The invention relates to a method for analyzing and / or processing a fertilized egg.
When studying the pathological processes of various diseases, it is often necessary to conduct medical experiments with manipulations on the chicken embryo. This raises the need for open access to the embryo. The access hole can reach a diameter of 1.5 × 2.5 cm. To obtain a positive result at the end of embryogenesis and hatching of the chick, it is necessary to maintain egg homeostasis, as an element - the restoration of the integrity of the shell.
A known method of analysis and / or processing of a fertilized egg (RF patent for the invention No. 2543263, IPC АКК 45/00, published on 02.27.2015), which includes the stage of puncture and / or injection of the substance into the egg white sack, for which there are eggs in the shell pierce a hole near the sharp end of the egg and insert a hollow needle into the hole. The distal end of the cannula is inserted into the protein bag without the needle passing through the bags with embryonic fluids or vital organs of the embryo and the substance of the protein bag is punctured and / or the processing substance is injected into the protein bag.
In this method, a small hole is made without touching bags with embryonic fluids or vital organs of the embryo. Therefore, to maintain egg homeostasis, there is no need to recreate the integrity of the shell. For experiments requiring large sizes of the access hole, this method is not applicable.
Closest to the proposed is a method of conducting an experiment on a chicken embryo (https://studopedia.ru/2_122933_metodi-eksperimentalnogo-zarazheniya-kurinih-embrionov.html) by infection of the embryo.
To infect the embryo with this method, it is placed in a tripod horizontally with the embryo up.
Two holes are made in the shell: one small above the center of the air chamber, and the other with a diameter of 0.2-0.5 cm laterally, from the side of the embryo. Infectious fluid is applied through the lateral opening to the surface of the chorionallantoic membrane and the opening is closed with a piece of adhesive plaster.
With this method, the problem of the forced position of the egg arises - since the adhesive patch gets wet and there is a risk of bacterial contamination of the covering material, since the impregnation occurs with protein, which is a good substrate for the growth of the bacterial flora. Also, the "forced" position of the egg - mainly in the vertical position - negatively affects the development of the embryo - the formation of deformities. Loss of chorioallantoic fluid due to soaking of the adhesive tape has a negative effect on the preservation of egg homeostasis. The inability to sterilize the adhesive patch significantly increases the risk of attaching the bacterial flora through the access "window". Due to the presence of adhesive content on the surface of the adhesive patch in contact with the protein part of the egg, there is a risk of peeling.
The objective (technical result) of the present invention is to create a method for analysis and / or processing of a fertilized egg, free from the above disadvantages.
The problem is solved in that the method of processing and / or analysis of a fertilized egg includes placing the egg horizontally with the embryo up, making an opening in the shell on the side of the embryo, performing analysis and / or processing of the embryo, and closing the opening in the shell. According to the invention, a three-dimensional model of a shell fragment is 8-15% larger than the size of a future hole using preliminary graphic information using computer graphics methods. According to the three-dimensional model, a mold is made in which a lid is molded from two-component silicone for injection molds to close the future hole. After making a hole in the shell and analyzing and / or processing the embryo, close the hole in the shell by applying a silicone cover to the hole and smoothing it from the hole to the edges.
3.As a two-component silicone, a high molecular weight organosilicon compound with the chemical formula [R 2 SiO] n , where R = an organic ethyl group of CH 2 —CH 3, can be used.
For the convenience of manipulating visual information using computer graphics methods, it is advisable to additionally form a three-dimensional model of the shell fragment from the side opposite from the future hole. In this case, according to the three-dimensional model, a mold is made in which the substrate for the egg is molded from polylactide, the egg is placed on the substrate, and then a hole is made in the shell on the side of the embryo, the processing and / or analysis of the embryo is performed, and the hole in the shell is closed.
The proposed combination of features of the proposed method ensures the preservation of homeostasis, allows to achieve the physiological completion of chick embryogenesis. The applied method allows for long-term observation. All this completely eliminates the risk of bacterial contamination when closing the access defect (the possibility of sterilizing the silicone cap by air at 180 ° C, with hot steam in an autoclave at 120-130 ° C, as well as the possibility of prolonged boiling in water). The possibility of egg rotation remains, eliminating the loss of chorioallantoic fluid.
3.The proposed method is as follows. According to visual information using computer graphics methods, a three-dimensional model of a shell fragment is formed 8-15% larger than the size of the future hole. According to a three-dimensional model, a mold is made in which a lid 1 is cast from a two-component silicone for injection molds to close the future hole. As a two-component silicone, a high molecular weight organosilicon compound with the chemical formula [R 2 SiO] n , where R = an organic ethyl group of CH 2 —CH 3, can be used.
The organic ethyl group (CH 2 -CH 3 ) in the composition of this compound - two-component silicone - is non-toxic to biological media while retaining the properties of silicone (inertness), thereby achieving long-term monitoring of the chicken embryo, without disturbing the physiological properties of the chorioallantoic fluid.
1).Based on the visual information using computer graphics methods, a three-dimensional model of the shell fragment is additionally formed from the side opposite from the future hole. According to a three-dimensional model, a mold is made in which
