Toxicity was studied on white mice (females) weighing 16-18 g with a single intraperitoneal administration in the form of a suspension in 2% starch mucus. The animals were monitored for 10 days, recording the behavior, intensity and nature of motor activity, the presence and nature of seizures, coordination of movements, skeletal muscle tone, reactions to tactile, sound and light stimuli, the frequency and depth of respiratory movements, heart rate, state hair and skin, the color of visible mucous membranes, water and food intake, weight change. For test compound 1, LD 50 is> 1200 mg / kg.
- 1998. -№1. - С. 27-32].Acute toxicity was calculated, following the recommendations of the state pharmacological committee for the study of the general toxic effects of biologically active substances [Methodological recommendations for the study of the general toxic effects of pharmacological agents. Approved 25.12.97 / Vestn. Pharmacopoeia. committee. - 1998.-№1. - S. 27-32].
М., 1977. - с. 196].According to the classification of toxicity of drugs, compounds I belong to class V of practically non-toxic drugs [Izmerov N.F., Sanotsky IV, Sidorov K.K. Toxicometry parameters of industrial poisons in a single exposure: Reference. M., 1977 .-- p. 196].
Antifungal activity was determined by the method of double serial dilutions in a liquid nutrient medium Saburo [Guidelines for preclinical studies of drugs. - Part 1. - M .: Grif and K, 2012. - S. 576-578]. For the test compound (I), the minimum inhibitory concentrations (MIC) were determined for the pharmacopeia strains of C. albicans ATCC 885-653, C. albicans ATCC 10231 obtained from the Scientific Research Institute of Medical Mycology named after P.N. Kashkina Federal State Budgetary Educational Establishment of the North-West State Medical University named after I.I. Mechnikov Ministry of Health of Russia, as well as in relation to clinical strains of C. krusei (1), C. krusei (2). Crops were made in Saburo liquid medium with various concentrations of test compounds. The studied compounds in an amount of 0.05 g were dissolved in 5 ml of dimethyl sulfoxide, 1 ml of the resulting dilution of 1: 100 was combined with 4 ml of Saburo's liquid medium. Next, a series of serial dilutions of compounds with a doubling concentration was prepared.
The cultures were grown in test tubes on a beveled agar medium (dense Saburo medium). A 48-hour culture was used to determine antifungal activity. To prepare a working suspension of microorganisms, the grown culture was washed off with an isotonic sodium chloride solution and the microbial suspension density was set according to the turbidity standard of 5 units. Next, a working solution with a concentration of 5 × 10 5 CFU / ml was prepared from the obtained microbial suspension. This suspension was added in an amount of 0.1 ml in test tubes with serial dilutions of the studied compounds. Thus, the microbial load in determining antifungal activity was 2-2.5 × 10 4 CFU / ml.
The results were taken into account after 48 hours of exposure of control and experimental tubes in a thermostat at a temperature of 24 ° C ± 1 ° C. The minimum inhibitory concentration (MIC) was determined by the absence of growth signs on the nutrient medium: the last tube with growth inhibition (transparent) corresponds to the MIC of the compound in relation to this strain.
Фунгистатический эффект исследуемого соединения I сравнивали с действием флуконазола [Celeste De Monte, Simone Carradori, Bruna Bizzarri, Adriana Bolasco et al. / Anti-Candida activity and cytotoxicity of a large library of new N-substituted-1,3-thiazolidin-4-one derivatives // European Journal of Medicinal Chemistry 2016, 107, P. 82-96].
