Abstract
Description
The invention relates to agricultural microbiology, biotechnology, the microbiological industry and agriculture, and in particular to microbiological means of optimizing the nitrogen nutrition of plants by pre-sowing seed treatment with effective diazotrophic bacteria and colonization of the roots.
И.А. В. - М., 2005. - 154 с.).The increase in crop yield is carried out by applying mineral and organic fertilizers. In modern biologization-oriented agriculture, one of the ways to increase crop yields, including rice, is the use of microbial preparations based on active strains of various microorganisms (Biological products in agriculture. Methodology and practice of using microorganisms in crop production and fodder production / Ed. I.A. Tikhonovich and Yu.V. Kruglov. - M., 2005. - 154 p.).
- 2012. - С. 11-16).In cereal crops, the preparation rhizoagrin based on the bacterium Agrobacterium radiobacter 204 with a set of plant-beneficial properties is used. (Kursanova BC, Bartaya NN “The effect of the preparation“ Rizoagrin ”on the yield of green mass of barley in single-seed crops and grass mixtures with legumes” / Bulletin of Altai State University No. 12 (122), 2012, S. 5-9; Melnichuk T .N. "The effectiveness of the use of biological products in the technology of cultivation of grain crops" / T.N. Melnichuk, N.K. Sherstoboev, I.A. Kameneva, L.A. Tchaikovskaya, S.S. Yaroshenko, V.V. Klyuchenko, N .M. Donetsk // Production Laboratory, No. 1 (40). - 2012. - P. 11-16).
№8).A known strain of rhizospheric bacteria Bacillus spp. KR-083, used as a means to protect agricultural plants from phytopathogenic microorganisms and stimulate their growth. The strain was isolated from the roots of rice growing in the Iengnam province of the Republic of South Korea and deposited under registration number "148 VNI-ISKHM-D" dated July 14, 2005 (RF Patent No. 2295562 C1, C12N 1/20, A01N 63/00, publ. March 20, 2007, bull. No. 8).
The lack of effectiveness of existing microbial preparations in rice cultivation is explained by the fact that intensive technologies are used in permanent crops of rice of modern varieties (repeated top dressing with nitrogen fertilizers, chemical treatment of plants with phytopathogens, phytophages and weeds). Such conditions form a microbocenosis that does not contribute to the survival of strains of their bioagents and, accordingly, to the functioning of the associative system of bacteria with plants.
The basis of the invention is the task of isolating a bacterial strain with a high degree of associativity with a plant adapted to the soil and climatic conditions of southern Russia and zoned rice varieties that can bind atmospheric nitrogen, stimulate plant growth and development, which will increase the crop yield.
The problem was solved by isolating in 2011 from the apical part of the roots of rice grown in a monoculture, a strain of associative bacteria Phyllobacterium ifriqiyense 6, an active nitrogen fixator, growth stimulant that increases rice productivity and can be recommended for the preparation of bacterial preparations.
The strain was isolated according to an improved method proposed by Sherstoboev N.K., Melnichuk T.N. and others, based on the principle of electivity, formulated by Vinogradsky, and the method of studying epiphytes according to Khudyakov (Krasilnikov N.A. Microorganisms of the soil and higher plants. M: Publishing House of the USSR Academy of Sciences, 1958. - 181 pp.) It was isolated and identified by its morphological and biochemical properties in 2011 according to Bergi's 1984 determinant and methodological recommendations for identifying non-spore bacteria that dominate the plant rhizosphere (Voznyakovskaya, Yu.M., Popova Zh.P., 1985). Genetic identification was carried out in 2016 at the Federal State Budget Scientific Institution All-Russian Agricultural Research Institute of St. Petersburg (with the support of grant No. 16-34-50060. Identification and characterization of the symbiotic properties of growth-promoting bacteria of the rhizosphere of rice).
6,9-7,2.The strain Phyllobacterium ifriqiyense 6 was isolated from the apical part of the roots of Khazar rice, which was grown in modified Leonard vessels on cabbage medium No. 19. Cultural-morphological and physiological-biochemical features of the strain: colonies on an agarized medium of rounded shape, with a diameter of 5-7 mm, convex, shiny, off-white (transparent), the edges are even. Cells are mobile, thickened sticks. Gram-negative, do not form a dispute. Abundant growth was noted on media with both mineral and organic nitrogen; utilizes simple saccharides with acidification of the medium (weak growth); starch does not hydrolyze, does not thin the gelatin; catalase positive; at 37 ° C no growth; optional aerob. Strain Phyllobacterium ifriqiyense 6 grows abundantly on a potato jamb, forming mucus. When cultivated in litmus milk, it peptones it. The temperature range of the growth of the strain 22-30 ° C. The optimum temperature is 25-28 ° C and a pH of 6.9-7.2.
Based on the analysis of the nucleotide sequence of the 16S rRNA gene, the newly isolated strain was identified as Phyllobacterium ifriqiyense and deposited in the Departmental collection of useful microorganisms for agricultural purposes (FGBNU VNIISKHM, St. Petersburg) under registration number RCAM 04327 dated 10.21.2016 (a copy of the certificate of deposit is attached )
