The invention relates to veterinary medicine, in particular to veterinary therapy, and can be used to stimulate regenerative processes in the cat's body - in the postoperative period, with degenerative diseases of organs and organ systems, acute and chronic skin diseases, improves the quality of life of aging animals.
State of the art
Known synthetic analogues having regenerative activity, such as sodium nucleinate, methyluracil and others. Such drugs have restorative properties, improve hemapoiesis, however, they have a relatively low effectiveness and narrow action, with a number of contraindications associated with the risk of depression of the nervous system, the occurrence of bradycardia, exacerbation of oncological pathologies of the blood system, and the occurrence of allergic reactions.
There is a regenerative drug “Laurabolin 50” (other names - retabolil, nandrolone decanoate for cats), which is an injectable anabolic steroid (nandrolone laurate), which is used as a general strengthening agent in the treatment of therapeutic and surgical diseases with significant tissue damage. The disadvantage of this drug is its steroid nature, which, in combination with a long course of treatment, can have a negative, dangerous effect on the whole body (especially the kidneys, liver and brain) and adversely affect the synthesis of the animal's own hormones.
Also known is the homeopathic preparation of systemic effects on the plant-based organism “Liarsin”, which consists of three components - plant raw materials Lycopodium and mineral compounds - Arsenicum and Phosphorus. Liarsin is used to restore the water-salt balance of cats after intestinal infections or inflammatory diseases of the gastrointestinal tract. The disadvantages of the known funds include the low biological activity of the drug and the limited scope, since it is used only for diseases of the gastrointestinal tract.
The prototype of the invention is the drug "Gamavit" - a complex preparation, the basis of which is the placenta, denatured emulsified and sodium nucleinate. The drug is available in liquid form on the basis of a growth medium containing a balanced solution of salts, amino acids and vitamins [RF Patent No. 20055475 C1 01.10.91]. For this prototype, the following effects on the body of cats are described: normalizes metabolic processes, has biostimulating, detoxifying, immunomodulating and adaptogenic effects.
The specified prototype has several disadvantages. One of the main components of the drug - the placenta - may carry a risk of developing an allergic reaction. The specified raw materials used to obtain this drug are not standardized, therefore, there is a danger of developing undesirable microflora, which entails the risk of infection of animals when using the drug. Also, the use of "Gamavit" is long-term and requires the introduction of a large amount of the drug (for prophylaxis at a dose of 0.1 ml per 1 kg of body weight, during treatment - 0.3-0.5 ml for 2-4 weeks). In addition, the drug is available in liquid form, which significantly reduces its shelf life.
The strategy of regenerative medicine, based on stimulating one's own regenerative potential, is used to create medicines for humans, for example, a conditioned environment that has a therapeutic effect in thermal burns [RU patent No. 2292212, 01/27/07], but is not used in veterinary medicine.
SUMMARY OF THE INVENTION
The present invention aims at providing stimulation of regenerative processes in the cat's body with a wide range of degenerative diseases and expanding the arsenal of drugs used in reconstructive veterinary medicine for cats.
The technical result is achieved by developing a veterinary preparation for cats with regenerative activity, and as a result, a therapeutic effect in relation to a wide range of diseases of cats.
The pathogenesis of degenerative diseases is associated with a violation of the ability of individual body tissues to recover and compensate by realizing their own regenerative potential. Existing prototypes have a pharmacological effect on the stimulation or replacement of the functions of preserved cellular elements. However, the concept of compensation for impaired activity and damage to target organs due to the indicated adaptive processes in a significant number of cases is untenable.
МСК присутствуют во всех органах взрослого организма и могут быть выделены и культивированы in vitro.MSCs have the ability to secrete a wide range of chemokines and growth factors and to respond quickly to changes in tissue homeostasis by modifying their secretion profile. Thus, with the participation of mesenchymal stem cells, a natural mechanism for regulating tissue regeneration is implemented. MSCs are present in all organs of the adult body and can be isolated and cultured in vitro.
The implementation of this approach can be achieved by using a preparation based on the extract and the conditioned culture medium of MSCs of bone marrow and adipose tissue, the regenerative activity of which is due to the action of a complex of cytokines secreted by MSCs in the culture medium.
The invention is an extract and a conditioned medium (CS) of a mesenchymal stem cell (MSC) culture (both de novo isolated and deconserved) from a bone marrow and / or adipose tissue of cats.
The manufacture of the claimed drug is carried out in the following steps:
1. The accumulation of cell mass and the formation of a conditioned environment. For this, deconservation of previously obtained or direct isolation of mesenchymal stem cells from the brain or adipose tissue of cats is performed. MSCs of cats are cultured in DMEM-based growth medium with 10% fetal bovine serum (FBS) until a conditioned medium is formed, the acceptability of which is assessed by the content of interleukins (IL-1β concentration of at least 2 pg / ml, IL-2 of at least 2 pg / ml, IL-6 - at least 10 pg / ml IL-10 - 2 pg / ml).
2. Preparation of the intermediate product on the basis of the conditioned medium and MSC extract. At this stage, a conditioned medium is collected that meets the criteria for acceptability for interleukin concentration, removed from the substrate and dispersed a monolayer of mesenchymal stem cells, a cell extract is obtained by freezing and thawing a cell suspension, and the extract is combined with a conditioned medium obtained by culturing a cat MSC.
3. The manufacture of the final product. The intermediate obtained in the previous step is stabilized by adding glycine at a concentration of 20 mg per 1 ml of lyophilization solution, subjected to sterilizing filtration, and bottling under aseptic conditions and freeze-drying.

The possibility of carrying out the invention and the effectiveness of its application are illustrated by examples.
Example 1. A method of obtaining a preparation based on an extract and conditioned medium of an MSC culture (both de novo isolated and deconserved) from a cat's bone marrow.
Клетки выращивали в ростовой среде, в состав которой входит питательная среда DMEM high glucose (Biowest, USA), L-glutamine, 10% фетальной бычьей сыворотки (ФБС, Sigma Aldrich, USA), гентамицин (80 мкг/мл), амфотерицин Б (25 мкг/мл, Biowest, USA).Cellular material in the form of bone marrow cells (CM) was obtained by puncture of the cat ileum using local anesthesia with 0.5% novocaine solution under strictly sterile conditions, 0.5 ml in volume. KM cells after isolation were placed in a chilled DMEM / F12 medium (Biowest, USA) containing gentamicin (80 μg / ml, Biolot, Russia) and amphotericin B (0.25 μg / ml, Biowest, USA), then washed twice from the blood DMEM / F12 medium with antibiotic and antimycotic by centrifugation at 1200 rpm for 5 minutes Cells were seeded at a concentration of 1 × 10 6 cells / ml and grown in plastic culture bottles (SPL, Korea) with a growth surface area of 175 cm 2 at 37 ° C in an atmosphere with 5% CO 2 content. Cells were grown in growth medium, which includes DMEM high glucose growth medium (Biowest, USA), L-glutamine, 10% fetal bovine serum (PBS, Sigma Aldrich, USA), gentamicin (80 μg / ml), amphotericin B ( 25 μg / ml, Biowest, USA).
To obtain a conditioned medium, the culture was carried out for 3-4 passages, then inoculation was carried out in culture bottles at the rate of 5-15 × 10 3 cells per 1 cm 2 of the surface area in 90 ± 5 ml of growth medium and cultivated under the conditions described above until the necessary concentration of interleukins in the environment.
Upon reaching the required concentration of biologically active substances, a conditioned medium containing all MSC secretion products was collected in sterile centrifuge tubes, purified by centrifugation at 3000 rpm and a temperature of 5 ± 1 ° C for 10 minutes from cell debris.
The resulting conditioned medium can be used directly for its intended purpose or stored for a long time under certain conditions (up to seven days at a temperature of 3 ± 1 ° C, longer storage requires a temperature of no higher than minus 20 ° C).
After collecting the conditioned medium, an extract of MSC KM was obtained: a cell monolayer, with a confluency of at least 90%, was disaggregated with the above dispersing solution, 0.9% sodium chloride solution (5-10 ml per 1 culture bottle) was added, it was frozen at a temperature minus 20 ± 2 ° C, after which thawing was performed at room temperature. The obtained MSC KM extract was collected in sterile centrifuge tubes, purified from cell debris by centrifugation at 3000 rpm and a temperature of 5 ± 3 ° C for 10 minutes.
To obtain a regenerative preparation, the centrifugation pellet, which is an extract of MSC KM, was combined with a conditioned medium, glycine was added (to a final concentration of 20 mg / ml), 1 ml was poured into vials, and freeze-dried.
So, by the end of the fourth week from the beginning of the cultivation of MSC KM cats, you can get up to 3000 ml (at a seeding concentration at the stage of isolation of at least 1 × 10 6 cells / ml) of a lyophilization solution based on the extract and conditioned medium of MSC KM, which can be used for the preparation of a drug that stimulates the regenerative processes of the cat's body.
Example 2. The specific activity of the drug based on the extract and the conditioned medium of the culture of MSCs (both de novo isolated and deconserved) from the bone marrow and / or adipose tissue of cats, in an in vitro model.
For this, the primary dermal fibroblasts of cats were taken at
At the end of the incubation, the grown cultures were removed from the plastic surface and the proliferative index was determined. The proliferation index (IP) was determined as the ratio of the number of cells in 1 ml of the suspension obtained by removing cells from the substrate to achieve confluence of the monolayer to the inoculum cell concentration (the number of cells in ml of the solution when plated on culture plates).
Data were obtained (Table 1 and Graph 1), which indicate an increase in the proliferative properties of model culture cells when a regenerative preparation based on the extract and conditioned MSC medium (both de novo and deconserved) was added to the growth medium % depending on the concentration in which the studied drug was added to the growth medium, compared with the control, while there was a linear relationship between the concentration of the drug and the value of the proliferation index.
Example 3. The study of the hepatoprotective properties of the drug based on the extract and the conditioned medium of the culture of MSCs (both de novo isolated and deconserved) of bone marrow and / or adipose tissue of cats.
The regenerative effect of the drug based on the extract and the conditioned culture medium of mesenchymal stem cells of bone marrow or adipose tissue was studied in rats in vivo acute liver injury (MOPP) model. The model of acute liver damage was performed by a single injection of rats into the stomach through a probe of paracetamol (N- (4-hydroxyphenyl) acetamide) in the form of a suspension at a dose of 700 mg per 1 kg of body weight. The treatment procedure after MOPP consisted in daily intramuscular administration of 0.5 ml of the studied drug to the animals of the experimental group for 5 days. As a control, rats with MOPP that were not subjected to therapeutic effects were used.
A day after the end of the course of treatment, the rats were removed from the experiment by decapitation, having previously provided anesthesia due to the intraperitoneal administration of chloral hydrate at a dose of 400 mg per 1 kg of animal body weight. In the blood serum, the activity of the aspartate- (AcT) and alanine aminotransferase (ALT) enzymes was determined using a standard Vital kit and the total protein content was determined using the biuret method. The studied biochemical parameters reflect the functional state of the liver. The enzymes AcT and Alt are present in significant quantities in the liver and kidneys, therefore, normally, the concentrations of AcT and Alt in the blood are low. When hepatocytes are damaged, the enzymes AcT and Alt are lost, and the concentration of these biochemical markers in the blood increases. A change in the level of total protein (a sign of gross pathology of the liver) was not observed.
2 и графики 2-4).According to the results of the study, it was shown that the use of a regenerative drug based on the extract and conditioned MSC medium of both bone marrow and adipose tissue and deconserved for five days in animals with AKI showed a significant improvement, judging by the studied markers of liver damage (ACT levels , ALT) in the group of animals treated with the regenerative preparation did not significantly differ from those in the group of intact rats (Table 2 and graphs 2-4).
Thus, a regenerative preparation based on the extract and the conditioned medium of MSCs has a pronounced therapeutic effect in acute damage to the liver of animals. Significant differences in the effectiveness of the drug based on the conditioned medium and the extract of mesenchymal stem cells obtained from various sources were also not detected.
Example 4. The effect of a regenerative preparation based on the extract and conditioned MSC culture medium (both de novo isolated and deconserved) of bone marrow and / or adipose tissue on wound healing in an in vivo model of a mechanical wound in mice.
Another model used to study the regenerative activity of the drug in vivo is a model of a deep mechanical wound, the edges of which were sewn to a plastic ring to prevent contraction. The drug was administered at the earliest stages of the wound healing process (during the first five days after modeling the wound) as subcutaneous injections. The study was carried out on 30 white outbred mice (15 mice — the control group (without treatment), 15 mice — the experimental group, which was used to pin the studied drug).
Five days after modeling the wounds, the animals of the experimental group did not have fibrin in the wound, exudation and hyperemia of the surrounding tissues were weak, swelling of the edges of the wound was observed, which indicated the subsidence of inflammation. An earlier shift of the peak of the phase of inflammation in the experimental group is most likely due to the action of growth factors and cytokines contained in the study drug. At the end of the proliferation phase, on the 14th day from the start of the experiment, the control group observed the ongoing process of epithelization of the wound surface, while the animals of the experimental group noted complete wound healing and signs of tissue remodeling, which indicated a higher rate of regeneration when using the studied drug.
Example 5. The study of the abnormal toxicity of the invention.
Tests for abnormal toxicity of the studied drug were carried out on white mice weighing from 19 to 21 g. The drug, heated to 37 ° C, was administered intraperitoneally in an amount of 0.5 ml to 20 mice. Monitoring the health of animals was carried out for 7 days. It was believed that the sample withstands the test if no animals die during the observation period.
The claimed preparation did not cause signs of toxicity or other side effects in animals.
Example 6. Clinical studies of a regenerative preparation based on an extract and conditioned medium of a culture of mesenchymal stem cells (both de novo isolated and deconserved) from bone marrow and / or adipose tissue of cats with hepatosis.
According to the invention, the claimed drug was administered to 17 cats with signs of hepatosis of different ages and sex belonging to private individuals.
The inventive drug was administered to cats in 0.2 ml, previously dissolving the contents of the vial in 1.0 ml of saline per 1 kg of animal weight intramuscularly daily for 5 days.
Sick animals were clinically monitored. To assess the state of health, blood samples were taken for laboratory tests on the day of admission, on the third, sixth and twelfth day from the start of therapy.
Clinical observation drew attention to the general condition of the animals, a change in appetite, the presence of vomiting, a change in fluid intake, the condition of the skin and mucous membranes (yellowness), and a macroscopic assessment of physiological excrement was performed.
An improvement in the general condition of animals was noted with a decrease or disappearance of icteric coloration of the skin and visible mucous membranes, dyspeptic symptoms, a decrease in pain during palpation of the liver, normalization of the general clinical and physiological state of cats, and the appearance of positive dynamics in biochemical studies of blood serum samples.
An analysis of the clinical data obtained in the treatment of cats revealed the following: by the third day that had passed since the start of the treatment with the claimed drug, an improvement in general condition occurred in eleven animals. They became more active, appetite appeared, thirst in three cats disappeared. No particular dynamics of clinical symptoms were observed.
By the sixth day, the general condition improved in all seventeen animals, in nine cats the yellowness of the visible mucous membranes significantly decreased, urine brightened, the stool assumed its usual natural color.
3.By 9-12 days, fourteen cats recovered, in the remaining animals, yellowness decreased. By 12-14 days, all animals recovered. The data obtained are presented in table. 3.
From the data presented in table. 4, it is obvious that by the sixth day of observation in cats, indicators of ALT and AcT activity, serum bilirubin concentrations returned to normal, which indicates reparative processes in the liver of these animals. Normalization of ESR and leukocyte concentration in animals indicate a weakening of the inflammatory process in the liver by this day. By the 12th day of the disease, all blood counts returned to normal on the background of the therapy. At the same time, it can be noted that by day 12, the glucose and total protein indicators, which when animals were admitted were at the level of the lower limit of the norm (which also indicates violations in the liver), reached average normal values.
Thus, the claimed drug shows a therapeutic effect in all observed cases of use, has pronounced regenerative, tonic, anti-inflammatory, immunostimulating activity and is intended for therapy in the postoperative period, to improve the living standards of aging animals, as well as for the treatment of degenerative diseases (liver cirrhosis, pulmonary fibrosis etc.), acute and chronic skin diseases.
Recommendations for the use of the proposed drug
The claimed drug is recommended to be administered by intramuscular or intravenous injection once a day for 3-7 days daily.
