Abstract
Description
The invention relates to the field of instrumentation mainly to measuring equipment. It can be used, for example, in materials science in studying the chemical composition of samples with nanometer spatial resolution, in the phonon spectrum of samples under study, in studying the patterns of behavior of nanostructures on the surface with the aim of further developing nanodevices.
[Optical Fluorescence Microscopy: From the Spectral to the Nano Dimension Summary, ed. By A. Diaspro, Springer, 2011; Лакович Дж. 1986. 488].A known method of researching objects, called fluorescence microscopy. In the framework of this method, some elements of interest to the researcher (molecules, cells, etc.) are marked with specific fluorescent labels. Next, the object is illuminated by a laser beam, causing these labels to fluoresce. After that, an image of the object under study can be constructed using fluorescent labels, which shows the spatial distribution of the elements of interest to the researcher and how this distribution changes over time. Fluorescence time, anisotropy, spectra and fluorescence kinetics can also be investigated, and more sophisticated fluorescence analysis methods can be used [Optical Fluorescence Microscopy: From the Spectral to the Nano Dimension Summary, ed. By A. Diaspro, Springer, 2011; Lakovich J. Fundamentals of fluorescence spectroscopy. 1986. 488].
The disadvantages of this method of research using fluorescence include:
- низкое разрешение без использования сверхразрешения;- low resolution without the use of super-resolution;
- невозможность управлять положением флуоресцентной метки при использования сверхразрешения.- the inability to control the position of the fluorescent label when using superresolution.
The closest analogue in technical essence and the achieved result is the method of excitation and registration of optical phonons in the cantilever needle of an atomic force microscope (AFM), which allows you to excite and register optical phonons on the tip of the AFM cantilever needle directly near the surface under study, and due to close mechanical connection the tip of the cantilever needle with the test surface to study the spectra of optical phonons of the test sample.
The disadvantages of this method of excitation and registration of optical phonons in the AFM cantilever needle include:
- довольно сложная экспериментальная техника, предполагающая использование фемтосекундного лазера;- A rather complex experimental technique involving the use of a femtosecond laser;
- ограниченный функционал, позволяющий экспериментатору работать только с оптическими фононами.- limited functionality that allows the experimenter to work only with optical phonons.
The technical result of the invention is to simplify the experimental technique used, on the one hand, and to increase the possibilities for studying physical phenomena on the surface with nanometer spatial resolution (chemical composition, viscoelastic properties, dielectric constant, etc.), on the other hand.
The specified technical result is achieved by the fact that the method of studying the surface using an atomic force microscope using fluorescence quantum dots, including laser irradiation of the active layer of the material deposited on the tip of a cantilever needle pressed against the sample, excitation and registration of optical phonons in it, processing the received information , differs in that as a layer of material deposited on the tip of the cantilever needle, there is a quantum dot interacting with the surface under study at which the exciting electrons using a laser pulse, then using the recording apparatus, focused on the quantum dot is recorded fluorescent emission, which displayed a quantum dot, is treated with the obtained information, and concludes properties of the medium on the basis of the fluorescent emission characteristics, which is a quantum dot.
In contrast to the closest analogue, one can use quantum dots of various compositions and devices, which will make it possible to study not only the spectrum of optical phonons, but also other surface properties that are accessible by fluorescence spectroscopy methods. In addition, a femtosecond laser is not required in experiments.
The nature of the fluorescence emission depends on the immediate environment of the quantum dot fixed on the tip of the cantilever needle.
A distinctive feature of the proposed method is the use of fluorescence radiation generated by a quantum dot, mounted on the tip of the cantilever needle, after its excitation by a laser beam. Due to the mechanical contact between the quantum dot and the surface under study, the properties of fluorescent radiation depend on the properties of the surface. Due to the fact that the quantum dot is located on the tip of the cantilever needle, it becomes possible to accurately position its placement on the surface under study.

7.A diagram of a device for studying a surface on an AFM using fluorescence quantum dots is shown in Fig. 1. The device includes a
A method for studying the surface on an AFM using fluorescent quantum dots is implemented as follows (Fig. 1).
Obtaining fluorescence emission spectra (as well as studying its other characteristics, such as fluorescence time, anisotropy, spectra and kinetics, etc.), combined with a 3D image of a probe microscope, will answer many pressing questions of condensed matter physics, chemistry, biology, medicine.
