Description
Изобретение относится к области медицины, а именно к офтальмологии, патофизиологии и экспериментальной трансплантологии, и может быть использовано для получения ткани хороидально-пигментного комплекса (ХПК) донора-трупа при снижении уровня загрязнения получаемой органной культуры ХПК фрагментами сетчатки и содержимым витреальной полости. Собственно сосудистая оболочка (ССО) глаза и располагающийся с ее внутренней стороны слой ретинального пигментного эпителия (РПЭ) вместе составляют ХПК и выполняют функцию двусторонней доставки многочисленных метаболитов из кровотока к слою фоторецепторов сетчатки и обратно, обеспечивая нормальную фототрансдуктивную функцию сетчатки. Патология РПЭ считается патогенетическим звеном дистрофических заболеваний сетчатки - возрастной макулярной дегенерации, пигментного ретинита, наследственных дистрофий сетчатки. При экспериментальной разработке способов лазерного и хирургического лечения указанных состояний традиционно используют модели экспериментальных животных, чаще всего кроликов. Данный подход имеет недостатки - по многим параметрам глаз животного отличается от глаза человека, эксперименты на животных относительно дороги и трудозатратны, требуют специально оборудованного вивария. В качестве альтернативы может быть использован метод органных культур, который позволяет получить небольшой жизнеспособный фрагмент интересующей ткани от трупного донора и поддерживать его жизнеспособность в течение некоторого срока. In the experimental development of methods for laser and surgical treatment of these conditions, models of experimental animals, most often rabbits, are traditionally used. This approach has drawbacks - in many respects, the animal’s eye is different from the human eye, animal experiments are relatively expensive and labor-intensive, require a specially equipped vivarium. As an alternative, the organ culture method can be used, which allows you to get a small viable fragment of the tissue of interest from a cadaveric donor and maintain its viability for some time. In particular, in some cases, it is possible to use the organ culture of COD to study the options for laser and surgical effects on the native RPE layer.
