Description of the invention
The aim of the present invention is to expand the arsenal of tools to reduce the dose of morphine hydrochloride while maintaining its analgesic effect, which helps to reduce the risk of morphine dependence. The authors of the present invention have found that the lack of an analog can be overcome by using the N-vinylpyrrolidone-based copolymer disclosed in RU 2415876 as a potentiating analgesic effect of morphine hydrochloride. For this copolymer, this type of activity is new and not obvious from the prior art.

где мономерное звено М представляет фрагмент 2-метил-5-винилтетразола (МВТ) или 2-метил-5-винилпиридина (МВП):where the monomer unit M represents a fragment of 2-methyl-5-vinyltetrazole (MBT) or 2-methyl-5-vinylpyridine (MVP):
и содержание мономерных звеньев n составляет 25-90 мол.%, а средневязкостная молекулярная масса Мµ сополимера зависит от природы М: если М представляет МВТ, то Мµ=100-250 кДа; если М представляет МВП, то Мµ=46-150 кДа в дозе от 200 до 2000 мкг/кг в качестве средства, потенцирующего анальгетический эффект морфина гидрохлорида.and the content of monomer units n is 25-90 mol.%, and the average viscosity molecular weight M µ of the copolymer depends on the nature of M: if M represents MW, then M µ = 100-250 kDa; if M represents the MEP, then M µ = 46-150 kDa in a dose of 200 to 2000 µg / kg as a means of potentiating the analgesic effect of morphine hydrochloride.
The ability to achieve the technical result of the invention, which consists in reducing the dose of morphine hydrochloride necessary to achieve a given level of analgesia, is confirmed in a series of generally accepted studies in animal models.
Example 1. The study of increasing the threshold of pain sensitivity (PBC)
Modeling of pain sensitivity is carried out by immersing the tail of the rat in water heated to 56 ° C. For the value of the threshold of pain sensitivity take the period of time from the moment the tail is immersed in water until it is removed from the water.
1.Wistar rats (males) of an initial weight of 100-120 g are placed in individual cells under conditions of immobilization of the body of the animal. After adaptation to the experimental conditions, the value of the threshold of pain sensitivity is measured in animals for 30 minutes, which is taken as 100% and is considered as a background indicator. Then the rats are randomly divided into groups of 7 animals each. Using blind control, the animals of the studied groups (Gy) are injected intraperitoneally with the claimed compound in various doses from 200 to 2000 μg / kg, and the animals of the control group - 0.9% NaCl solution in the same volume. Then, in animals of all groups for 90 minutes with an interval of 15 minutes, pain threshold parameters are measured (group average ± s.o.), which are expressed as a percentage in relation to the background value. The test results are presented in table 1.
As follows from table 1, with a single peripheral injection of the copolymer in accordance with the invention in the entire dose range, the threshold values of pain sensitivity in animals do not change, which indicates the absence of an analgesic effect in the copolymer.
Example 2. The study of changes in the threshold of pain sensitivity with the simultaneous introduction of a copolymer and morphine hydrochloride
