Abstract
Description
The invention relates to the field of plant physiology, in particular to the regulation of the accumulation of ascorbic acid in plants.
The work shows the stimulating effect of an electric field of longitudinal orientation on the content of the reduced form of ascorbic acid (AA) in eight-day-old barley seedlings.
It is known that the synthesis of ascorbic acid increases in plants in the light [1], under the action of certain chemical compounds and biotechnological products [2], however, the effect of the electric field on this process has not been studied.
The well-known "Method of stimulating the synthesis of ascorbic acid (vitamin C) in barley seedlings" according to the patent of the Russian Federation No. 2102869, A01G 31/00, decl. 1995.06.06, publ. 1998.01.27, which consists in the following: cut leaves of seedlings are placed by slice in an aqueous solution of dioxiacetone in a concentration of 0.1-1.0% and illuminated with fluorescent lamps for 24 hours.
The disadvantages of this method can, in our opinion, include, firstly, the use of dioxiacetone as a chemical stimulant, and secondly, the insufficiently high accumulation of ascorbic acid.
The aim of the proposed method is to stimulate the synthesis of ascorbic acid in a more environmentally friendly way: by exposure to an electric field of longitudinal orientation on lighted leaves equipped with an exogenous substrate.
This goal is achieved due to the fact that the cut leaves are placed with the base in a 1% glucose solution, a substrate for AK biosynthesis, in the electric field with an intensity of 5 × 10 5 V / m with the direction of the tension vector from the top of the sheet to its base.
The use of 1% glucose solution as a substrate contributes to the production of environmentally friendly crop products. Placing plants in an electric field with a certain intensity when the vector is directed from the top of the sheet to the base allows one to significantly intensify the process of AA synthesis.
An example of the method
2×c.The process of stimulation of AA in plants was carried out in an installation that consists of two power supplies: low voltage with a voltage of ± 15 V, high voltage - 25 kV and a flat capacitor, between the plates of which barley leaves are placed. The installation is located in the TKSh flora - 1 chamber with fluorescent lamps with an intensity of 12.5 J / m 2 × s.
The effect of the field on the AK system of barley seedlings is calculated by the following formula:
где C(+) - концентрация АК в опытных растениях,where C (+) is the concentration of AK in experimental plants,
C (-) is the concentration of AK in control plants.
The possibility of stimulating the synthesis of ascorbic acid by an electric field is demonstrated by the experimental data presented in the drawing.
The drawing shows that after two hours of plants in an electric field, stimulation of the synthesis of ascorbic acid begins, which reaches its maximum value after 3 hours. In the future, the stimulation effect decreases and completely disappears with an 8-hour exposure.
Given the above data, we came to the conclusion that it is advisable to stimulate the synthesis of ascorbic acid in plants by placing them for three hours in an electric field of longitudinal orientation with a voltage of 5 × 10 5 V / m with the direction of the tension vector from the top of the sheet to its base.
2×c.In this case, the leaves should be provided with an exogenous substrate for the biosynthesis of ascorbic acid - 1% glucose, and also illuminated with light intensity of 12.5 J / m 2 × s.
This method of increasing the level of ascorbic acid in plants can find application in obtaining high-quality, vitamin, environmentally friendly crop products.
