Abstract
Description
The invention relates to medicine, and specifically to the assessment of the hemostatic properties of topical agents to stop bleeding.
When developing a variety of hemostatic agents, it is possible to evaluate their performance and choose the optimal formulation by conducting a series of experiments on animals.
Existing methods for assessing the hemostatic activity of hemostatic agents in vitro are not adequate to the physiological processes occurring in the body. Modeling the process of hemostasis outside the body is always very arbitrary.
д.The well-known "Method for the detection and comparative evaluation of the hemostatic activity of hemostatic matrix preparations of local action" (Patent RU 2127428, IPC 7 G 01 N 33/48, N33 / 49, 03/10/1999, 93044526/14, 09.09.1993). It consists in the following: under anesthesia, the animal is operated on, providing access to internal organs, then a bleeding wound is applied and bleeding is stopped using a liquid stopping agent applied to a sponge, tampon, etc.
The disadvantages of this method are the duration of the study, the reliability of the results.
The technical result of the proposed method for evaluating the hemostatic effect of liquid hemostatic agents is to reduce the study time while increasing the reliability of the results.
The method is as follows:
In laboratory conditions, 1 ml of the patient’s blood, taken in a 1: 9 ratio, stabilized with a 3.8% sodium citrate solution, is taken after the coagulological tests necessary for the patient. Next, 0.1-0.2 ml of a liquid styptic is added dropwise and everything is mixed with a glass rod. Turn on the stopwatch and note the presence of a hemostatic effect when blood clots appear for no more than one minute.
Example No. 1: In 1 ml of the patient’s blood stabilized with a 3.8% sodium citrate solution, taken in the ratio 1: 9, after the coagulological analyzes necessary for the patient were carried out, 0.1 ml of MK-7M medical cyanoacrylate adhesive was introduced dropwise. It was not possible to mix the glue with blood, because it instantly polymerized on the surface of the blood upon contact. Conclusion: glue cannot stop active bleeding, which is observed in practice.
Example No. 2: In 1 ml of patient’s blood stabilized with a 3.8% sodium citrate solution, taken in a ratio of 1: 9, after the coagulological analyzes necessary for the patient were carried out, 0.2 ml of surgical gelatin-resorcinolum "Glue-Lab "and everyone mixed up. The formation of a clot within 1 min did not occur, however, the blood easily mixed with glue and within 3-5 min, as the glue polymerized, a uniform film formed. Conclusion: "Bioglue Lab" has a weak hemostatic effect.
Example No. 3: In 1 ml of the patient’s blood stabilized with a 3.8% sodium citrate solution, taken in a ratio of 1: 9, after the coagulation tests necessary for the patient were carried out, 0.1 ml of Kaprofer hemostatic agent was added dropwise and everything was mixed . The formation of a loose clot occurred within 15 s. Conclusion: Kaprofer has hemostatic activity, which is confirmed in practice.
The technical result is achieved in that the evaluation of the liquid hemostatic effect is carried out in laboratory conditions on stabilized human blood after coagulological analyzes.
The application of the method leads to a significant reduction in the time for evaluating the hemostatic effect, so the animal experiment lasts for several hours, and in the laboratory the result is obtained within a few minutes.
The biochemical composition of human blood is different from the biochemical composition of animal blood, therefore, a more accurate assessment of the hemostatic effect of hemostatic agents is carried out.
