Abstract
Description
The invention relates to medicine, experimental biology, ecology, toxicology and can be used to study the mechanisms of toxic effects of heavy metals, in particular cobalt, on kidney function.
Cobalt is an important biologically active element necessary for the normal implementation of vital processes - providing erythropoiesis, the synthesis of nucleic acids, vitamins and hormones, regulating the activity of enzymes and all types of metabolism.
А. Гончаревская и соавт., 1985; A. Jungwirth et al., 1990; Y.B. Zhang et al., 1998; и др.).The increasing use of cobalt and its compounds in industry in recent decades has contributed to the identification of toxic effects of cobalt on the human body (E. Szakmary et al., 2001), up to stillbirth and developmental anomalies. Excessive intake of cobalt in the body leads to its accumulation in the tissues, to the greatest extent of the liver and kidneys, since the main ways of excretion are secretion of bile and urination. The toxic effect of cobalt along with the widely studied selective myocardial damage in the form of the so-called cobalt cardiomyopathies (M.P. Chekunova, 1978; I.M.Suvorov et al., 1978; A.E. Vermel et al., 1991) is manifested by kidneys - the main site of cobalt excretion from the body and therefore the target organ for a damaging toxic effect. Under the action of excess cobalt on the body, dystrophic and necrobiotic changes in the renal epithelium are described (E. Szakmary et al., 2001), impaired renal tubule function and transmembrane transport of ions and water (O.A. Goncharevskaya et al., 1985; A. Jungwirth et al., 1990; YB Zhang et al., 1998; et al.).

According to the authors, the essence of the claimed invention should not be obvious for specialists from a known level of medicine, since the above possibility of obtaining a method for modeling toxic nephropathy is not revealed from it, which allows us to conclude that the criterion of “inventive step” is met.
The set of essential features characterizing the essence of the invention, in principle, can be repeatedly used in medicine to obtain a result consisting in a more accurate and easily reproducible method for producing chronic toxic nephropathy, which allows us to conclude that the invention meets the criterion of “industrial applicability”.
This method is as follows.
To obtain a toxic substance, cobalt chloride is dissolved in sterile distilled water in such a way that 0.4 mg of cobalt for a dosage of 4.0 mg / kg and 1.0 mg of cobalt for a dosage of 10.0 are obtained per unit of solution equal to 0.2 ml mg / kg (i.e. 4.0 mg / kg = 0.4 mg / 100 g / 0.2 ml of solution; 10.0 mg / kg = 1.0 mg / 100 g / 0.2 ml of solution) . For every 100 g of rat weight, 0.2 ml of a toxic solution is injected, which is not an excessive water load on the body of an experimental animal. A solution of cobalt chloride is introduced through a probe into the stomach, daily 1 time per day for two months.
Fifteen white rats male Wistar strain weighing 200-300 g through a probe into the stomach was injected with a solution of cobalt chloride at a dose of 10.0 mg / kg once a day for two months. Every 2 weeks, functional changes in the kidneys were observed: diuresis, glomerular filtration, tubular reabsorption of water, excretion of sodium, potassium and calcium. Comparisons were made with indicators of intact animals (control).
Выведение кальция с мочой было достоверно выше фона через 2 недели (р<0,001) и 1 месяц (р<0,01), а, приблизившись через 1,5 месяца к фоновому значению, к концу второго месяца значительно снижалась (р<0,001).Analyzing the data in Tables 1 and 2, it can be noted that the processes of urination after 1.5 months were significantly changed: glomerular filtration decreased (p <0.02), and tubular reabsorption of water increased (p <0.05), which led to a significant a drop in diuresis (p <0.01). After 2 months, the value of diuresis decreased even more (p <0.001), which was due to a further decrease in glomerular filtration (p <0.01) and an increase in tubular reabsorption of water (p <0.01). Sodium excretion significantly decreased (p <0.01) after 1.5 months, and by the end of 2 months it fell even more (p <0.001). Determination of potassium excretion after 2 weeks; 1; 1.5 and 2 months of the experiment did not reveal significant deviations from the background. Urinary calcium excretion was significantly higher than the background after 2 weeks (p <0.001) and 1 month (p <0.01), and, approaching the background value after 1.5 months, significantly decreased by the end of the second month (p <0.001) .
3.After 1 and 2 months of the experiment, the rats were sacrificed using thiopental to study kidney tissue. Determination of cobalt content in the tissue of the kidneys, urine and urinary excretion revealed a significant increase in the metal content after 1 and 2 months of the experiment, which is shown in Table 3.
For histological studies, samples of kidney tissue were fixed in 10% neutral formalin, and then subjected to paraffin filling, followed by preparation of sections 7-8 microns thick. Sections were stained with hematoxylin and eosin. The sections were studied in transmitted light using a Mikmed-1 microscope under a magnification of 80 × 200 × 400. Histological studies revealed necrotic nephrosis of the renal tubule epithelium, the structure of the tubules was not determined (figure 1, 2). Edema and accumulation of protein in the lumen of the tubules were pronounced.
Functional changes in renal tissue were confirmed by histological studies, which revealed a picture of toxic nephropathy.
Thus, the proposed method for modeling chronic toxic nephropathy shows the effectiveness of its use.
