Изучена возможность использования геля с метронидазолом для заживления резаных ран роговицы у крыс. Исследования ранозаживляющей активности геля с метронидазолом проводились на белых половозрелых крысах без учета пола массой 180-220 г, разделенных на три группы. В первой группе 6 животных, а в остальных группах по 4 экспериментальных животных в каждой. Под эфирным наркозом (с экспозицией 2-3 мин) произведено иссечение поверхностных слоев роговицы левого глаза крыс микрохирургическим лезвием (под микроскопическим контролем), на глубину до 1/2 толщины роговицы, в центральной ее части, площадью до 1/3 площади роговицы. В двух случаях произошла перфорация роговицы. Данные животные не были включены в общую статистику. В послеоперационном периоде ежедневно 2 раза в день произведено нанесение глазных мазей гелем 1% (первая группа), гелем метронидазола 0,5% (вторая группа), вазелиноланолиновой основой (третья группа). Выведение животных из эксперимента произведено на 3-е, 7-е и 14-е сутки. При выполнении экспериментально-морфологических исследований на первый срок выведения животных из эксперимента (3 сутки) во всех трех группах наблюдалась полная эпителизация дефекта роговицы. В связи с этим анализ морфологических данных проводили по нормализации структуры эпителия. Толщину эпителия оценивали по средним значениям вариационного ряда в микронах; наличие патологических разрастаний эпителия по качественным параметрам; присутствие “+”, отсутствие “-”; дифференцировку слоев эпителия по трехбалльной системе: один балл - отсутствие, два бала - слабая, три бала - полная (таблица). При выполнении экспериментально-морфологических исследований на третий срок выведения животных из эксперимента (14 сутки) в первой группе (гель 1%) наблюдали редукцию кровеносных капилляров, эпителий равномерный, под эпителием типичная базальная мембрана не сформирована. В строме равномерно распределены кератобласты, что соответствует норме, а толщина стромы составляет 100 мкм (фиг.1). Далее при выведении из эксперимента (14 сутки) во второй группе (гель с метронидазолом) структура всех слоев роговицы восстановлена, с толщиной стромы 90 мкм. Толщина эпителия 20-25 мкм, состоит из 5 слоев клеток, которые четко дифференцируются, равномерно распределены кератобласты, восстановлена базальная мембрана. Клетки базального слоя эпителия имеют округлые ядра, которые постепенно уплощаются к поверхностным слоям. Не нарушена и структурная целостность эндотелиальных клеток, т.е. идет полноценное восстановление эпителия (фиг.2). В третьей группе (вазелиноланолиновая основа) очевидно замедление регенерации, истонченная строма и невосстановляющаяся структура роговицы. Роговица расслоилась, видно эпителий и строму. При окраске по Ван-Гизону сохраняется неравномерность эпителия на протяжении дефекта, толщина стромы составляет 60 мкм (фиг.3).The possibility of using a gel with metronidazole for the healing of cut wounds of the cornea in rats was studied. The animals were removed from the experiment on the 3rd, 7th and 14th day. When performing experimental-morphological studies for the first term of removing animals from the experiment (3 days) in all three groups, complete corneal defect epithelization was observed. In this regard, the analysis of morphological data was carried out to normalize the structure of the epithelium. The thickness of the epithelium was estimated by the average values of the variation series in microns; the presence of pathological growths of the epithelium in qualitative parameters; presence of “+”, absence of “-”; differentiation of the layers of the epithelium according to a three-point system: one point - absence, two points - weak, three points - complete (table). When performing experimental-morphological studies for the third period of animal elimination from the experiment (14 days) in the first group (1% gel), reduction of blood capillaries was observed, the epithelium was uniform, a typical basement membrane was not formed under the epithelium. In the stroma, keratoblasts are evenly distributed, which corresponds to the norm, and the stroma thickness is 100 μm (Fig. 1). Then, when removing from the experiment (14 days) in the second group (gel with metronidazole), the structure of all layers of the cornea was restored, with a stroma thickness of 90 μm. The thickness of the epithelium is 20-25 microns, consists of 5 layers of cells that are clearly differentiated, keratoblasts are evenly distributed, and the basement membrane is restored. The cells of the basal layer of the epithelium have rounded nuclei, which gradually flatten to the surface layers. The structural integrity of endothelial cells, i.e. there is a full restoration of the epithelium (figure 2). In the third group (the petrolatum base), a slowdown in regeneration, a thinned stroma and a non-healing structure of the cornea are evident. The cornea exfoliated, visible epithelium and stroma. When stained according to Van Gieson, the epithelium is not uniform throughout the defect, the stroma thickness is 60 μm (Fig. 3).

Это дает возможность врачу в практической медицине, в зависимости от формы и тяжести заболевания рекомендовать больным нанесение глазного геля на конъюнктиву самостоятельно, что делает заявляемый гель перспективной лекарственной формой в офтальмологической практике. После проведения полномасштабных клинических испытаний для лечения заболеваний глаз - кератитов (акантомебных, герпетических), конъюнктивитов и разработки методик контроля качества планируется оформление фармакопейной статьи.Observation on patients showed a positive effect of the eye gel, and the amount of drug applied at this ratio of components is the same.
