Abstract
Description
The invention relates to the field of medical industry, in particular to the production of an inducer virus for the production of human leukocyte interferon.
In the production of human leukocyte interferon, it is envisaged to use the vaccine strain "H" of the Newcastle disease virus (VBI) as an inducer virus of interferon. This strain is used in many countries around the world that produce human leukocyte interferon for medical and veterinary practice or in scientific research. This is the safest inducer virus with inducing properties in the production of human or animal leukocyte interferon.
A known method of producing the virus-inducer of interferon by culturing strain "H" of the Newcastle disease virus in 9-11 day old chicken embryos [1]. The infectious dose of the virus is 10 6 CPD / 50 / 0.2 ml. The inducer virus is diluted in 0.1 M phosphate-buffered saline. This dose is injected into the allantoic cavity of the embryo with a syringe subject to sterile conditions. Then the embryos are incubated for 48 hours at (32 ± 0.5) o C. After incubation, the embryos are cooled at 4 o C and the virus-containing allantoic fluid is aspirated, which, after necessary controls, is used as the interferon inducer virus. Virus-containing fluid must meet the following requirements: be bacteriologically sterile, have a hemagglutinating activity (GA) of 256 units, an infectious titer of at least 10 11 CPD / 50 / ml. Using this inducer in the production of human leukocyte interferon, interferon with an antiviral activity of 11,000 to 14,000 IU / ml is obtained.
The objective of the invention is to increase the interferon-inducing ability of a viral inducer.
To solve the problem, in the method for producing the interferon inducer virus, the uterine material (strain H) of the Newcastle disease virus GKB N 2348) is diluted with phosphate-buffered saline with the addition of 10.0% D (+) - lactose, introduced into the allantoic cavity of chicken embryos concentration of 10 6 JRC / 50 / 0.2 ml, followed by incubation and selection of virus-containing fluid.
A distinctive feature of the proposed method is the addition of D (+) - lactose in phosphate-buffered saline during the dilution of the uterine strain, which provides higher titers of interferon with an activity of 16,000 - 18,000 IU / ml versus 11,000 - 14,000 IU / ml in the prototype (see table).
Example 1. The strain "N" of the virus of Newcastle disease (HBV No. 2348) was used as a uterine strain. To obtain a production strain, a 9-10 day old chicken embryo is introduced into the allantoic cavity under sterile conditions at a concentration of 10 6 CPD / 50 / 0.2 ml of the initial suspension of the mother strain, diluted in phosphate-buffered saline, with the addition of 10.0% D ( +) - lactose. Incubated for 48 hours at a temperature of (32 ± 0.5) o C. After incubation, the embryos are cooled for 18-20 hours at 4 o C, the virus-containing allantoic fluid is suctioned off. The α-type interferon titer obtained with this inducer is 16,000 IU / ml.
Example 2. The strain "N" of the virus of Newcastle disease (HBV No. 2348) was used as a uterine strain. Incubated for 48 hours at a temperature of (32 ± 0.5) o C. After incubation, the embryos are cooled for 18-20 hours at 4 o C, the virus-containing allantoic fluid is suctioned off. The α-type interferon titer obtained with this inducer is 18,000 IU / ml.
