Изобретение относится к ветеринарной вирусологии, а именно к способу приготовления инактивированной культуральной вакцины для профилактики чумы крупного рогатого скота (КРС). Известен способ приготовления инактивированной вакцины из лимфоузлов и легких больных чумой КРГ телят, зараженных вирулентным вирусом чумы КРС (Базылев П. М. докт.дисс. М. 1955). В настоящее время инактивированные вакцины против чумы КРС отсутствуют (Сергеев В.А. в кн. "Вирусные вакцины", Киев, 1993). Для профилактики чумы КРС в нашей стране и за рубежом используют живые вирусвакцины. (А.с. N 194262 от 30.06.65 г.). Для получения вируса по известному способу используют первичнотрипсинизированную культуру клеток почки теленка (ПТ), выращенную на 0,5% гидролизате лактальбумина в матрасах вместимостью 1-1,5 дм3. Активность получаемого вируса составляет 105,0-106,25 ТЦД50/см3. Указанная цель достигается тем, что используют вакцинный штамм ЛТ (а.с. 174765 от 15.07.65), выращенный в культуре перевиваемой линии клеток почки сайгаков (ПС), а в качестве инактиватора вируса сернокислую медь. Круговой монослой культуры клеток заражают вакцинным штаммом ЛТ вируса чумы КРС в дозе 0,1-1,0 ТЦД50. На клетку и выращивание вируса проводят в роллерном аппарате при скорости вращения сосудов 10-12 об/час, при температуре 36-38oС в течение 3-4 суток. При развитии характерных для вируса цитопатических изменений и поражении 80-90 клеток, содержимое сосудов встряхивают для отделения клеток от стекла и сливают в общую стерильную емкость.
