Abstract
Description
The invention relates to methods for the isolation of biologically active plant substances of the pharmaceutical industry and technical entomology, in particular to a technology for producing individual ecdysteroids from a mixture thereof.
378-382; Volodin V.V., Alexeeva L.I., Kolegova N.A. "Further ecdysteroids of Serratula coronata L.// Biochemical systemat. and ecology.- 1998.- V.26.- issue 4.- P. 459-461). The mixture of ecdysteroids from the aerial part of Serratula coronata consists of 20-hydroxyecdysone (75%), inocosterone (11%), ecdysone (5%), as well as macisterones A and C, augosterone C, dacrychainansterone, which are present in trace amounts (Volodin B.V. ., Luksha V.G., Dainan L. et al. "Inocosterone and Macisterone A from Serratula coronata L. // Plant Physiology .- 1998.- V. 45.- N 3.- S. 378-382; Volodin VV , Alexeeva LI, Kolegova NA "Further ecdysteroids of Serratula coronata L. // Biochemical systemat. And ecology.- 1998.- V.26.- issue 4.- P. 459-461).
М. Д. Ф. A known method of isolating ecdysone from a mixture of ecdysteroids obtained from plants of the genus Serratula coronata, which is carried out by applying a mixture of ecdysteroids to a chromatographic column with aluminum oxide, followed by elution with a mixture of organic solvents, evaporation and recrystallization (a.c. CCCP 1146050. Method for producing β-ecdysone from a plant of the genus Serratula // A.M. Grodzinsky, Yu.D. Kholodova, I.F. Mitunin, etc.).
The disadvantages of this method are the low productivity of the process due to the use of large volumes of solvents and the duration of the process.
(Alekseeva L. , Volodin V., Luksha V. "A convenient method for ecdysteroids concentration// XIII-th Ecdysone Workshop.- Jena.- July 27-31, 1998: Programme and abstracts.- P. 16). There is also a method of producing 20-hydroxyecdysone and ecdysone from a mixture of ecdysteroids, adopted as a prototype. The method is based on continuous elution of an aqueous solution of a mixture of ecdysteroids through a hydrophobic sorbent until the concentration of ecdysone in the solution at the outlet and in the initial solution is equalized (Alekseeva L., Volodin V., Luksha V. "A convenient method for ecdysteroids concentration // XIII-th Ecdysone Workshop .- Jena.- July 27-31, 1998: Program and abstracts.- P. 16).
The disadvantage of this method is the length of the process due to the low solubility of 20-hydroxyecdysone in water.
The objective of the invention is to increase the productivity of the process, reducing productivity and energy consumption of the process.
Distinctive essential features of the invention is the use of aqueous solutions of organic solvents as an eluent for applying ecdysteroids to the sorbent, which increases the solubility of ecdysteroids by at least 5 times and, thus, leads to a significant decrease in the volume of solution passed through the sorbent, reducing the duration of the process, and therefore makes the process less time consuming.
The method is as follows:
A mixture of ecdysteroids obtained from the aerial part of a plant of the genus Serratula coronata is dissolved in an aqueous-organic solution (1-25%). The resulting solution of ecdysteroids is continuously passed through a layer of hydrophobic sorbent (according to the prototype) until reaching the maximum capacity for ecdysone, which has a greater affinity for the surface of the sorbent than 20-hydroxyecdysone, inocosterone and macisterone. The achievement of the maximum capacity for ecdysone is controlled by thin layer chromatography (TLC) or HPLC. Then, a 25-100% aqueous solution of organic solvents is eluted through the sorbent layer. As organic solvents, methanol, ethanol, isopropanol, n-butanol, a mixture of ethyl acetate and methanol, etc. are used. The solvent is evaporated, and the concentrate enriched with ecdysone thus obtained is purified by preparative HPLC and recrystallization.
The proposed method of enrichment of the ecdysteroid mixture with minor components simplifies their further isolation by known methods of preparative of-HPLC.
An example implementation of the method.
