Abstract
Description
The invention relates to methods for the isolation of individual aromatic and heterocyclic amino acids from mixtures thereof and can be used in the chemical, microbiological, food industry, and agriculture.
Known methods for the separation of amino acids, which are based on classical ion-exchange chromatography [A.S. USSR 644782, C 07 C 99/12, 1979, A. p. USSR 639862, C 07 C 99/12, 1978, A.S. USSR 979991, G 01 N 31/08, 1981, A. p. USSR 960163, C 07 C 99/12, 1982]. The disadvantages of these methods are the use for the regeneration of a significant amount of auxiliary reagents, the presence of the stage of preliminary preparation of amino acid solutions before passing through the ion exchanger, a long time for the separation process. Closest to this invention is a method for the separation of proline with valine (leucine, hydroxyproline) [RF Patent 2144530 from 16.07.97. ]. The disadvantages of this method is the length of the process associated with the low rate of flow of the mobile phase through the layer of a chromatographic carrier to establish sorption equilibrium and the need for regeneration of the sorbent after each separation cycle.
The objective of the proposed method is to increase the efficiency of separation of a mixture of phenylalanine and tryptophan with a significant reduction in the duration of the process and the exclusion of an additional stage of regeneration of the ion exchanger.
The problem is achieved by passing a mixture of phenylalanine and tryptophan through a column with anion exchange resin AB-17-2P [Ionites (catalog). Cherkasy: Publishing House of NIITEKHIM, 1975. 38 pp.] In the OH form at a rate of 6 ml / min, eluting with 0.1 M NaOH solution and fractional separation of phenylalanine and tryptophan solutions at the column outlet.
The use of 0.1 M NaOH solution as an eluent eliminates the need for sorbent regeneration, since the release of amino acids proceeds with the simultaneous conversion of the ion exchanger into the OH form. The preference for choosing the indicated flow rate of the mobile phase through the chromatographic support layer is due to the existence of a region of dynamic sorption modes in which an increase in the transmission rate of the eluent leads to an improvement in the separation of the components of the mixture. The maximum separation of amino acids is observed at an eluent transmission rate of 6 ml / min. In this case, the resolution of the components is increased by 2.4 times compared with the equilibrium mode of the process (transmission rate of 2 ml / min) and the separation cycle time is reduced by 3 times. A further increase in the feed rate of the mobile phase leads to a deterioration of separation, since the output volumes of the components approach the value of the free volume of the column.
Ч.1.295 с. 4.2. 621 с.]).Example. The separation is carried out at a temperature of 20 o C on a column with highly basic anion exchange resin AB-17-2P in OH-form weighing 3.0000 g; the height of the sorbent layer is 12 cm. An artificially prepared mixture is introduced into the column, containing 0.3020 g of phenylalanine and 0.1825 g of analytical grade tryptophan dissolved in 50 ml of a 0.01 M NaOH solution. A 0.1 M NaOH solution was used as eluent. 25 ml fractions are collected at the column exit, which are analyzed for the content of shared amino acids spectrophotometrically on a Specord M 40 quartz cuvette 1 = 1 cm at 257 nm for phenylalanine and 279 nm for tryptophan, taking into account the effect of amino acids on each other's analytical signal. The maximum separation of amino acids is observed at an eluent transmission rate of 6 ml / min. Moreover, the resolution of the components increases by 2.4 times compared with the equilibrium mode of the experiment (transmission rate of 2 ml / min) and the separation cycle time is reduced by a factor of 3. The first 75 ml of the resulting amino acid-free solution from the column is discarded. The next 300 ml, not containing tryptophan and containing phenylalanine at a concentration of 0.402 g / l, (which is 40% of the initial content of this amino acid in the mixture) is collected. Then, 500 ml of the solution resulting from the column contain both phenylalanine 0.360 g / l (60% of the content in the mixture) and tryptophan 0.166 g / l (45% of the content in the mixture) and are sent to the next separation cycle. A further 950 ml, not containing phenylalanine and containing tryptophan at a concentration of 0.106 g / L (55%), was collected. Thus, as a result of the separation cycle, three fractions are obtained: the first contains only phenylalanine (0.1206 g) without tryptophan impurity; the second is a mixture of amino acids; the third contains only tryptophan (0.101 g) without phenylalanine impurities. The degree of purity of the obtained individual amino acids reaches 95% (control was carried out by TLC according to [Sharshunova M., Schwartz V., Mikhalets Ch. Thin-layer chromatography in pharmacy and clinical biochemistry. M: Mir, 1980. Part 1.295 p. 4.2. 621 s .]).
